β-Escin Effectively Modulates HUVECs Proliferation and Tube Formation

Abstract: In the present study we evaluated the anti-angiogenic activities of β-escin (the major active compound of Aesculus hippocastanum L. seeds). Human umbilical-vein endothelial cells (HUVECs) were used as an in vitro model for studying the molecular mechanism underlying the anti-angiogenic effect of β-escin. We investigated the in vitro effects on proliferation, migration, and tube formation of HUVECs and in vivo anti-angiogenic activity was evaluated in a chick chorioallantoic membrane (CAM) angiogenesis assay. “…The CAM assay was performed as described in a previous study (Varinská et al. 2018 ). Chicken eggs at seven days post-fertilization (Boehringer Ingelheim, Beijing) were cleaned with 70% ethanol, and a window of ∼1.5–2.0 cm 2 was gently opened on the blunt end of the egg without damaging the embryo.…”

“…The effect of escin on the invasiveness of NCI-H23 cells was determined by Boyden chamber assay [26]. Escin treated NCI-H23 cells were seeded at the density of 1×10 5 Gelatin zymography in the collagen-coated upper chamber of the Boyden chamber (8 μm polycarbonate membrane). The bottom compartments of the Boyden chamber were filled with a 10% complete medium.…”

“…Escin is a saponin derived from the Aesculus hippocastanum. Several research studies have evidenced that anti-oxidant [2], anti-cancer [3], anti-inflammation [4], antiangiogenic properties [5], reverses multidrug resistance [6]. Escin has the ability to reduce MDA-MB-231, and KBM-5 cells invasion by regulation of invasive genes such as extracellular matrix (ECM) degradation enzyme matrix metalloproteinase-9 (MMP-9) [7][8][9].…”

“…Qualitative and quantitative analysis of HCE sample was realized by HPLC-DAD-MS according to previously reported method [23]. Briefly, all HPLC analysis were carried out on the Agilent Technologies 1200 Series chromatograph (Agilent Technologies Deutschland, Walbron, Germany), equipped with a diode array spectrometer and MS ion trap detectors (AB SCIEX 3200 Q TRAP LC/MS/MS System, Applied Biosystems, Foster City, CA, USA).…”
