β‑elemene induced anticancer effect in bladder cancer through upregulation of PTEN and suppression of AKT phosphorylation

Abstract: Human bladder cancer is one of the most aggressive tumours known and has shown resistance to traditional chemotherapy, which depends heavily on DNA-damaging drugs. β-elemene is one of the least cytotoxic antitumor agents that are extracted from Curcuma aromatica salisb and it exhibits antitumor effects in many carcinomas. β-elemene has attracted the attention of clinicians and scientists worldwide due to its few side effects and limited effect on the bone marrow. However, the antitumor mechanism of β-elemene r… Show more

“…2) is a sesquiterpene mixture isolated from various Chinese herbs such as Curcuma wenyujin Y. H. Chen et C. Ling, Rhizoma zedoariae, and Curcuma Zedoary. It has various pharmacological effects including anti-oxidative, anti-inflammatory and anti-cancer activities [744][745][746]. It exerts anti-cancer effects in many cancers, such as lung, gastric, cervical, breast and bladder cancers, osteosarcoma, through apoptosis, inhibition of cell proliferation, migration and invasion, angiogenesis [746][747][748][749][750][751][752].…”

“…β-Elemene is shown to induce apoptosis in human cervical cancer SiHa, NSCLC A549 cells, primary bladder cancer cells, and Burkitt's lumphoma, and inhibit tumor growth in Lewis tumor-bearing mice [746,747,749,753,754]. It up-regulates insulin-like growth factorbinding protein 1 (IGFBP1) to induce a reciprocal interaction between microRNA 155-5p and FoxO3a, which leads to the inhibition of cell growth in human NSCLC A549 and H1975 cells [755].…”

“…Previously, researchers reported that β-elemene prevents the migration of various cancer cells, such as cells of bladder cancer, cells of melanoma, cells of lung cancer, cells of stomach cancer, cells of cervical cancer and cells of breast cancer [ 15 , 23 , 24 , 25 , 26 , 27 ]. In our study, using a wound healing assay, we discovered that β-elemene inhibits the migration of human glioblastoma cells.…”

“…However, β-elemene is present not only in Cannabis sativa but also from Curcuma rhizome, and it is commonly used in traditional Chinese medicine due to its anticancer properties with no reported severe side effects [180]. In this way, this compound has been extensively studied as an anticancer agent in vitro and in vivo and has been demonstrated to be a promising drug for the treatment of a wide variety of tumors [181][182][183][184][185][186]. Among the challenges associated to cancer treatment, it is the development of multidrug resistance (MDR), which negatively impacts the effect of chemotherapy drugs, and consequently treatment success.…”
