Bacillus anthracis secretes three polypeptides: protective antigen (PA), lethal factor (LF), and edema factor (EF), which interact at the surface of mammalian cells to form toxic complexes. LF and EF are enzymes that target substrates within the cytosol; PA provides a heptameric pore to facilitate LF and EF transport into the cytosol. Other than administration of antibiotics shortly after exposure, there is currently no approved effective treatment for inhalational anthrax. Here we demonstrate an approach to disabling the toxin: high-affinity blockage of the PA pore by a rationally designed low-molecular weight compound that prevents LF and EF entry into cells. Guided by the sevenfold symmetry and predominantly negative charge of the PA pore, we synthesized small cyclic molecules of sevenfold symmetry, ␤-cyclodextrins chemically modified to add seven positive charges. By channel reconstitution and high-resolution conductance recording, we show that per-6-(3-aminopropylthio)-␤-cyclodextrin interacts strongly with the PA pore lumen, blocking PA-induced transport at subnanomolar concentrations (in 0.1 M KCl). The compound protected RAW 264.7 mouse macrophages from cytotoxicity of anthrax lethal toxin ‫؍(‬ PA ؉ LF). More importantly, it completely protected the highly susceptible Fischer F344 rats from lethal toxin. We anticipate that this approach will serve as the basis for a structure-directed drug discovery program to find new and effective treatments for anthrax.infectious diseases ͉ membrane transport ͉ modified cyclodextrins

A ciklodextrinek gyûrû alakú szénhidrátmolekulák, melyek három méretben (6, 7 és 8 tagú alfa-, béta-és gamma-ciklodextrin) megtalálhatók a természetben is. A növekvõ igények miatt ma már több tízezer tonnát gyártanak évente a természetes ciklodextrinekbõl, és egyre nagyobb mennyiséget a legfontosabb származékokból, így a hidroxipropil-, random metil-és szulfobutil-béta-ciklodextrinbõl (HPBCD, RAMEB és SBEBCD) is.Az elmúlt csaknem 40 év alatt a ciklodextrin (CD) technológia a korszerû ipar szinte minden területén talált gyakorlati alkalmazást. A ciklodextrin-tartalmú termékekbõl jelenleg több mint 60 humán gyógyszer, 10 állatgyógyászati készítmény és több száz élelmiszeripari-, kozmetikai-és háztartás-vegyipari termék található a piacon. E termékek közös sajátsága, hogy bennük a ciklodextrinek, csupán segédanyagok, kapszulázó ágensek, a termékek minõségének javítására, egyes alkotóinak stabilizálására, vízoldékonyságuk, felszívódásuk, biológiai hasznosulásuk fokozására szolgálnak. Napjainkig a ciklodextrinek által kapszulázott hatóanyagok nagy része természetes és szintetikus kismolekula volt, ezek molekulatömege a 200-700 Dalton közötti tartományba esett.A korszerû biotechnológiai eljárásokkal sejt-és szövetkultúrákban termelt biológiai hatóanyagok mind molekuláris méretük, mind molekulatömegük tekintetében jelentõsen eltérnek a fent említett kis molekulás hatóanyagoktól. A terápiás hatású fehérjék molekulatömege már a 2000-300.000 Dalton tartományba esik. Ezen érzékeny makromolekulák sajátos, olykor kiszámíthatatlanul változó térszerkezete, molekulafelszíni polaritási viszonyaik miatt a ciklodextrinekkel történõ egyszerû "host-guest" (gazda-vendég) típusú kölcsönhatásuk tanulmányozása a szokásos analitikai módszerekkel nehezen végezhetõ. Nem meglepõ tehát, hogy a ciklodextrinek irodalma, a napjainkig közölt kb. 4500 fehérje-peptid tárgyú közlemény ellenére, a ciklodextrinek hatása tekintetében nem mutat egységes képet. Számos -olykor egymásnak ellentmondó -eredmény miatt is nehéz általános következtetéseket levonni a ciklodextrin-fehérje kölcsönhatás nyújtotta lehetõségekrõl. Ennek egyik feltételezhetõ oka, mely az irodalom tüzetes átnézésekor derül ki, hogy több, egyébként színvonalas közleményben, a szerzõk az alkalmazott ciklodextrinek típusát nem kellõen definiálták. Egyes szerzõk pl. béta-ciklodextrinnek nevezik a származékolatlan béta-ciklodextrint (helyesen), éppúgy, mint a metil-béta-ciklodextrint, a szulfobutiléter-béta-ciklodextrint és a 2-hidroxipropil-béta-ciklodextrint is. A ciklodextrinek és fehérjék kölcsönhatásával kapcsolatos közzétett eredmények tehát nem pontosan köthetõk egy bizonyos cilodextrin típus hatásához. We compared the abilities of structurally related cationic cyclodextrins to inhibit Bacillus anthracis lethal toxin and Staphylococcus aureus ␣-hemolysin. We found that both ␤-and ␥-cyclodextrin derivatives effectively inhibited anthrax toxin action by blocking the transmembrane oligomeric pores formed by the protective antigen (PA) subunit of the toxin, whereas ␣-cyclodextrins were ineffective. In contrast, ␣-hemolysin was selectively blocked only by ␤-cyclodextrin derivatives, demonstrating that both symmetry and size of the inhibitor and the pore are important.Previously, we proposed a novel approach for the discovery of inhibitors of pore-forming toxins that involves the blockage of the pores using molecules with comparable dimensions and the same symmetry as the target pores. It allows for the identification of lead compounds faster and significantly more cheaply in comparison with the existing industry standards. First, this approach was successfully tested on anthrax lethal toxin (LeTx), which plays a key role in anthrax infection. The toxin was disabled by the blockage of the pore formed by protective antigen (PA 63 ), an essential component of anthrax toxin, by rationally designed compounds. Based on the 7-fold symmetry of the PA 63 pore, we synthesized and tested cyclic molecules that had 7-fold symmetry using ␤-cyclodextrin (␤-CD) as a starting molecule (Fig. 1). The discovered inhibitors of anthrax toxin were successfully tested in vitro and in vivo (1-5, 7, 10). The broader applicability of this approach was demonstrated using as targets two other toxins that form transmembrane pores with 7-fold symmetry: ␣-hemolysin (␣-HL) of Staphylococcus aureus (2, 11) and ε-toxin produced by Clostridium perfringens (unpublished data).To investigate how the structural features of the pore blockers affect their activities, we evaluated the ability of structurally related derivatives of ␣-, ␤-, and ␥-cyclodextrins to inhibit the cytotoxic activities of anthrax lethal toxin (LeTx) and staphylococcal ␣-HL as well as to block the ion current through the channels formed by PA 63 and ␣-HL in planar lipid membranes.
