β Cell mass expansion during puberty involves serotonin signaling and determines glucose homeostasis in adulthood

Abstract: Puberty is associated with transient insulin resistance that normally recedes at the end of puberty; however, in overweight children insulin resistance persists leading to an increased risk of type 2 diabetes. The mechanisms whereby pancreatic b cells adapt to pubertal insulin resistance, and how they are affected by the metabolic status, have not been investigated. Here we show that puberty is associated with a transient increase in b-cell proliferation in rats and humans of both sexes. In rats, b-cell prolif… Show more

“…This may explain the inverse association between GHR and IGF-1 that we observed. Increased GHR signaling in pancreatic islets has been shown to regulate pubertal beta cell expansion in rats . However, little is known about the association of plasma GHR levels with systemic metabolism.…”

“…Increased GHR signaling in pancreatic islets has been shown to regulate pubertal beta cell expansion in rats. 35 However, little is known about the association of plasma GHR levels with systemic metabolism. Plasma GHR levels were recently reported to be associated with incident T2D in a large cohort study, 36 and reductions in GHR levels were identified as a causal mediator of improvements in glycemia after bariatric surgery in adults with T2D.…”

“…We show that CDX2 drives the expression of serotonin synthesis genes in these pre-β-cells. Serotonin production is a feature of neonatal and adolescent β-cells (Castell et al, 2022; Moon et al, 2020b), and adult β-cells can activate serotonin synthesis during pregnancy (Baeyens et al, 2016; Kim et al, 2010; Moon et al, 2020a; Ohara-Imaizumi et al, 2013). Based on this evidence, we posit that SC-ECs are not an erroneous intestinal cell type of SC-islet in vitro differentiation but rather represent a cell type characteristic of normal human β-cell development.…”

“…Pancreatic β-cells adhere to this pattern, featuring a complex system for controlling cell mass ( Bouwens and Rooman, 2005 ). Consequently, this endocrine organ is distinguished by the determination of developmental trajectories, influencing various aspects of feeding behavior and adaptive physiological traits in adult life ( Kim et al, 2010 ; Castell et al, 2022 ). Our discovery reveals that in sea urchins, the post-oral neurons is a part of a complex neuroendocrine regulator, resembling its operation in vertebrates.…”

“…However, the regulation of cell mass in pancreatic β-cells is still intertwined with these ancient monoaminergic regulatory systems. β-cells express 5-HT receptors, and 5-HT increases their proliferation in pubertal islets ( Castell et al, 2022 ). DA synthesis is absent in β-cells in vertebrates.…”
