β-adrenergic receptor signaling evokes the PKA-ASK axis in mature brown adipocytes

Abstract: 21Boosting energy expenditure by harnessing the activity of brown adipocytes is a promising 22 strategy for combatting the global epidemic of obesity. Many studies have revealed that the β3-adrenergic 23 receptor agonist is a potent activator of brown adipocytes, even in humans, and PKA and p38 MAPK have 24 been demonstrated for regulating the transcription of a wide range of critical genes such as Ucp1. We 25 previously revealed that the PKA-ASK1-p38 axis is activated in immature brown adipocytes and 26 contr… Show more

“…In LoxP mice, acute cold exposure (24 h) elicited only mild overall STK signaling responses (Fig.S3d), whereas chronic cold stimulation for 6 days decreased the activity of members of the CMGC kinase family (including cyclin-dependent kinases (CDK), glycogen synthase kinases (GSK) and MAP kinases (MAPK)), and activity of AGC family members (including protein kinase A (PKA), C (PKC), and G (PKG) were elevated (Fig.S3ei,ii). The CMGC family includes important hubs of cAMP-dependent signaling like the MAPKs p38 42 , ERK 43 , JNK 44 , which are typically engaged during acute thermogenic activation 45 . However, STK signatures in BAT of Adcy3-AdcKO at 22°C resembled chronic cold exposure in LoxP mice with exception that PKA activity remained unchanged (Fig.S3f, i-iii).…”
