α-Xylosidase plays essential roles in xyloglucan remodelling, maintenance of cell wall integrity, and seed germination inArabidopsis thaliana

Abstract: HighlightXyloglucan oligosaccharide metabolism by α-xylosidase impacts xyloglucan remodelling, the mechanical integrity of the primary cell wall of growing tissues, cell expansion, and seed germination.

“…The broad expression of XLY1 in the embryo and the endosperm during seed development and imbibition was shown in the former study using the XYL1 promoter (3 kb) (Sechet et al ). In contrast, the latter study showed that the promoter (∼2 kb) activity was enriched in the radicle tip and the upper hypocotyl of the embryo in 24 h‐imbibed seeds, with only low activity detected in the lower hypocotyl and the transition zone between the radicle tip and hypocotyl (Shigeyama et al ) (Figure A). The reason for this discrepancy is not known.…”

“…In the trg1‐1 ( xyl1 mutant) seeds imbibed at 34 °C, the expression of the two ABA biosynthesis genes ZEAXANTHIN EPOXYDASE ( ZEP ) and NINE‐CIS‐EPOXYCAROTENOID DIOXYGENASE9 ( NCED9 ) is reduced as compared to WT, resulting in a decrease in ABA levels. In contrast, the expression of GA3ox‐1 , a rate‐limiting GA biosynthesis gene, is enhanced in the trg1‐1 seeds imbibed at 22 °C (Shigeyama et al ). These results suggest that alteration of the cell wall properties could affect the ABA‐GA balance in seeds.…”

“…An α‐xylosidase TGR1/XYL1 has been identified that could finely tune the levels of xyloglucan oligosaccharides and their action on the mechanical properties of the CW. The expression of the TGR1/XYL1 gene is high in growing tissues such as the root tip and the upper hypocotyl, but relatively low in the elongating cells in the embryo of the germinating seed (Shigeyama et al ., ).…”
