Extensive application of pesticides is usually accompanied with serious problems of pollution and health hazards. Lambda-cyhalothrin (LCT), a type II synthetic pyrethroid, is widely used in agriculture, home pest control and protection of foodstuff. This study designed to evaluate the dose dependent haematological, hepatic and gonadal toxicity of LCT at different dose levels in Wistar rat. Investigations were also done to find out the toxic effect of lambda cyhalothrin on lipid metabolism in female rat and its amelioration by taurine. Rats were exposed to different doses of lambda cyhalothrin over a period of 14 consecutive days. Exposure to LCT produced ataxia, agitation, rolling and also tremors which were considered as the signs of toxicity. Significant decrease in erythrocyte count, haemoglobin percentage, seminal fructose concentration, hepatic and testicular reduced glutathione (GSH) content was observed. Increase in leukocyte count, serum aspartic and alanine transaminase, hepatic and testicular malondialdehyde (MDA), testicular and ovarian cholesterol after LCT treatment were seen in male rats at the dose level of 10.83 mg/kg body wt. (1/7 th LD 50 ). Elevated ovarian cholesterol and MDA and reduced 3β hydroxy steroid dehydrogenase (HSD) and GSH level were also observed in lambda cyhalothrin exposed female rat at the dose level of 6.29 mg/kg body wt.(1/9 th LD 50 ). LCT caused increase in serum triglyceride, cholesterol, low density lipoproteins (LDL), very low density lipoproteins (VLDL) and bilirubin and decrease in serum high density lipoproteins (HDL) in female rat. Taurine pretreatment ameliorated LCT induced altered lipid metabolic biomarkers in female rat.

Kidney is one of the most affected organs by aluminium toxicity. This study aimed to investigate the effect of aluminium chloride on the kidney of pregnant rats and to assess the efficiency of vitamin E and selenium in ameliorating this effect. Forty virgin albino rats were randomly divided into two main groups. Control rats were further divided into negative control group (C1, n = 10) which received distilled water and positive control group (C2, n = 10) that received vitamin E (VE, 150 mg/kg/day) and selenium (NaSe 150 µg/kg/day) for 3 months through intra-gastric tube. The experimental group was divided into an E1 subgroup in which rats received aluminium chloride (AlCl 3 , 150 mg/kg/day, n = 10) and E2 subgroup (n = 10) in which animals received the same dose of AlCl 3 plus VE and selenium at the same doses as C2 group for 3 months through intra-gastric tube. Conception of rats was allowed. AlCl 3 , VE and NaSe were given through intragastric tube during the whole length of the pregnancy, at the same doses as before pregnancy. At the 20th day of gestation dams were sacrificed, kidneys were dissected and processed for routine histological and immunohistochemical staining for identification of T-lymphocytes and macrophages. Integrated optical density of both cell types was assessed. AlCl3 administration induced histopathological changes in the kidney of pregnant rats and increased the density of CD3 and CD68 immunoreactive cells, suggestive of the associated aluminium-induced inflammatory process. Vitamin E and selenium minimized these harmful effects. The results suggest that diets rich in vitamin E and selenium and their supplements are advised particularly during pregnancy to alleviate the effects of possible excessive aluminium exposure.

Vitamin E (-Tocopherol [-Toc]) is the primary membrane bound, lipid-soluble, chain-breaking antioxidant that has been reported to protect against lipid peroxidation-induced tissue damage. Therefore, the present study was designed to investigate the protection afforded by α-Tocopherol (vitamin E), if any, on LPS-induced liver injury in Wistar rats and to study the underlying mechanism of protection. Bacterial endotoxin (10 mg/Kg body weight-prestandardized dose) was injected intraperitoneally and animals were sacrificed 8 h post-challenge. Vitamin E (35 mg/Kg body weight) was administered orally for 15 days prior to endotoxin challenge. Challenge with LPS resulted in a significant increase in the activities of serum transaminases and alkaline phosphatase along with histological alterations in the liver. These responses were associated with elevated levels of malondialdehyde (MDA) and reduced levels of glutathione, superoxide dismutase and catalase along with increased levels of tumor necrosis factor alpha (TNF-α) in the liver homogenates. However, vitamin E supplementation attenuated the oxidative stress by reducing the levels of MDA, restoring the levels of glutathione, superoxide dismutase and catalase and decreasing the levels of TNF-α. Decreased TNF-α levels after vitamin E supplementation might have resulted into the modulation of above mentioned biochemical changes resulting into amelioration of hepatic architecture. Vitamin E, therefore, seems to have a promising role for clinical manifestations due to oxidative stress.
