α2D/A‐adrenergic receptor gene induction in the retina by phorbol ester: involvement of an AP‐2 element

Abstract: Background: The ␣ 2 -adrenergic receptor (␣ 2 -AR) expressed in the bovine retina has been demonstrated to be of the ␣ 2D subtype. The bovine ␣ 2D -adrenergic receptor (␣ 2D/A -AR) gene has been cloned and characterized. This report describes the induction of this gene by phorbol-12, 13-myristate acetate (PMA), an activator of protein kinase C (PKC).

“…Alcohol is a known activator of PKC in various cell types[3235] and PKC has been implicated in the induction of the α 2A -AR gene transcription [36, 37]. As reported by Venkataraman et al ., [37] the increase in α 2A -AR mRNA resulting from the exposure of rat C6 glioma cells to phorbol- 12, 13-myristate acetate (PMA) was mediated by PKC through the AP-2 element of the α 2A -AR promoter. PMA, like alcohol, is an activator of PKC [38, 39].…”
