α 2 -Adrenoceptor-Mediated Hyperpolarization of Locus Coeruleus Neurons: Intracellular Studies in Vivo

Abstract: Intracellular recordings in vivo from noradrenergic neurons in the rat locus coeruleus showed that membrane potential was hyperpolarized by the administration of clonidine (an alpha 2-adrenoceptor agonist) or after a burst of spikes evoked by intracellular pulses; both types of hyperpolarization were associated with a decrease in membrane input resistance, and both could be blocked by the alpha 2-adrenoceptor antagonist piperoxane. These results suggest that a hyperpolarization of membrane potential mediated b… Show more

“…Reinstatement of drug seeking is also obtained with administration of the α-2 adrenoreceptor antagonist yohimbine, which, by increasing brain noradrenaline cell firing (Aghajanian and VanderMaelen 1982) and release (Abercrombie et al 1988), acts as a pharmacological stressor (Bremner et al 1996a,b;Charney et al 1983;Holmberg et al 1962;Lee et al 2004;Lê et al 2000Lê et al , 2005. These two methods were used to investigate the effect of URB597 on stress-induced alcohol seeking.…”

“…LC neurons fire spontaneous action potentials. Activation of ␣ 2A AR or -opioid receptors in the LC enhances outward K ϩ current through inwardly rectifying K ϩ channels, leading to neuronal hyperpolarization and consequent reduction in spontaneous firing frequency (37)(38)(39). As shown in Fig.…”

“…4 C and D). The attenuation of spontaneous firing in the LC is attributed primarily to activation of inwardly rectifying K ϩ channels (37)(38)(39), so the observation that the D79N ␣ 2A AR is unable to regulate LC neuronal firing suggests that the ␣ 2A AR cannot activate these K ϩ currents in D79N mice. Activation of ␣ 2 AR results in inhibition of voltage-gated Ca 2ϩ channels in many neuronal preparations.…”

“…First, burying behavior was evoked by yohimbine, which blocks α 2 autoreceptors to increase NE cell firing and release (Aghajanian and VanderMaelen 1982;. Second, administration of the non-selective ß-adrenergic antagonist propranolol significantly attenuated CRF-evoked burying, as well as grooming and head shakes, implicating the ß-adrenergic receptor in these behavioral responses.…”
