α-Amylase gene transcription in tissues of normal dog

Abstract: We studied the distribution of alpha-amylase mRNA in normal dog tissues by northern blotting (NB) and reverse transcription-polymerase chain reaction (RT-PCR) with human pancreatic (AMY2) and salivary (AMY1) alpha-amylase cDNA-specific primers. Analysis of poly(A+) RNA from various normal tissues by NB indicated the presence of detectable levels of alpha-amylase mRNA transcripts only in pancreas. Dot-blot analysis of DNA amplified with primers common to both (human) isoamylase mRNAs showed presence of alpha-am… Show more

“…Whether this is due to important differences in the populations of patients in previous human studies and dogs included in the present study or to a true diminished risk of HES-associated AKI in dogs compared to people is unclear. Explanations for possible interspecies differences may be a greater capacity to degrade HES in dogs due to greater ␣-amylase activity (the enzyme responsible for HES breakdown) 49 or diminished accumulation due to a higher rate of elimination compared to people, proven with high molecular weight HES (8 d vs 13 d, respectively). 45 In our study population, the median daily dose of HES administered was significantly lower than the maximal daily dose for people recommended by the manufacturers at the time of administration of the product (50 mL/kg/d).…”
