Zoledronic acid regulates the synthesis and secretion of IL-1β through Histone methylation in macrophages

Abstract: Long-term administration of nitrogen-containing bisphosphonates increases the risk of detrimental side effects, such as bisphosphonate-related osteonecrosis of the jaw (BRONJ). BRONJ development is associated with inflammation, but its pathophysiology remains unknown. Here, we examined whether histone methylation is responsible for zoledronic acid (Zol)-induced inflammatory responses. We found that Kdm6a and Kdm6b markedly increased interleukin 1β expression and Gasdermin D cleavage, which are both activated b… Show more

“…Zoledronic acid triggers M1-like macrophage polarization and increases the mRNA and protein levels of IL-1β by activating the NLRP3 inflammasome and cleaving pro-caspase-1 in LPS-primed THP-1 cells; these effects can be reversed by silencing of ASC ( 131 ). Zoledronic acid also induces caspase-1-dependent and GSDMD-mediated pyroptosis and secretion of IL-1β in RAW264.7 cells, by mediating methylation of histone H3 (H3k27me3) ( 193 ). Furthermore, increased numbers of Th17 cells and IL-17 levels were found to be correlated with elevation of the M1/M2 macrophage ratio in human and murine BRONJ lesions ( 302 ).…”

“…Moreover, the capacity of PC1 to prevent BTZ-induced increase of some proinflammatory cytokines, such as IL-1β, suggests that PK system might epigenetically regulate inflammatory processes involved in pain maintenance. In this regard, previous studies showed the KDM6A’s ability to regulate H3k27me3 demethylation at the IL-1β gene promoter [ 42 ].…”

“…Recent studies have revealed an essential role of macrophages in the development of BRONJ ( Zhu et al, 2019 ; Yang et al, 2020 ; Zhao et al, 2020 ). We previously showed that macrophages orchestrate inflammation trends by shifting their phenotypes in microenvironment, in which efferocytosis is necessary for its anti-inflammatory response ( Zhu et al, 2019 ).…”
