Longevity Assurance 5 (LASS5), a member of the LASS/Ceramide Synthases family, synthesizes C16-ceramide and is implicated in tumor biology. However, its precise role is not yet well understood. A yeast two-hybrid screen was performed using a human cDNA library to identify potential LASS5-interaction partners. One identified clone encodes succinate dehydrogenase subunit B (SDHB). Mammalian two-hybrid assays showed that LASS5 interacts with SDHB, and the result was also confirmed by GST pull-down and co-immunoprecipitation assays. The C-terminal fragment of SDHB was required for the interaction. LASS5 and SDHB were co-localized in COS-7 cells. LASS5 and SDHB expressions were found to be up-regulated in neuroglioma tissue. Transfection assays showed that LASS5 or SDHB expression repressed p53 or p21 reporter activity, respectively. Simultaneous LASS5 and SDHB expression resulted in stronger repression of p53 and p21 reporter activity, suggesting that LASS5 and SDHB interaction may synergistically affect transcriptional regulation of p53 and p21. Our data provide new molecular insights into potential roles of LASS5 and SDHB in tumor biology.

Studies of human asthma and of animal models of allergic airway inflammation revealed a crucial role for Th2 cells in the pathogenesis of allergic asthma. Kruppel-type zinc finger proteins are the largest family of a regulatory transcription factor for cellular development and function. Zinc finger protein (Zfp) 35 is an 18-zinc finger motif-containing Kruppel-type zinc finger protein, while its function remains largely unknown. The aim of this study was to clarify the role of Zfp35 in the pathogenesis of Th2-dependent allergic inflammation, such as allergic asthma. We examined airway eosinophilic inflammation and hyperresponsiveness in two mouse models, which use our newly generated Zfp35-deficient (Zfp35−/−) mice and adoptive transfer of cells. In Zfp35−/− mice, Th2 cell differentiation, Th2 cytokine production, eosinophilic inflammation, and airway hyperresponsiveness were substantially enhanced. Furthermore, adoptive transfer of Ag-sensitized Zfp35−/− CD4 T cells into the asthmatic mice resulted in enhanced airway inflammation and airway hyperresponsiveness. These results indicate that Zfp35 controls Th2 cell differentiation, allergic airway inflammation, and airway hyperresponsiveness in a negative manner. Thus, Zfp35 may control Th2-dependent diseases, such as allergic asthma.

Abstract. The data analysis of the on-board instruments set of the Cosmos-1809 and IKB-1300 satellites is presented for the following events. Continuous sequence of tropical cyclones (TCs) after the Mounts Pinatubo and Hudson eruptions in 1991. Sequences of 3 super-typhoons in the Western Pacific in November 1990. Sequences of 5 hurricanes in the Atlantic Ocean in September 1981. It is shown that the effects of the several interacting TCs impact on the ionosphere differ from those of single TCs. The distribution of electron fluctuations and density, VLF-ELF oscillations, and other plasma parameters of the upper ionosphere varies at different stages of development of tropical disturbances. Models of TCs interaction that change the parameters of the ionosphere are proposed. An important role is played by acoustic oscillations generated in TCs. The strongest volcanic eruptions in the twentieth century threw a significant amount of aerosols into the stratosphere. It is discussed how stratosphere changes for interacting TCs and perturbations after volcanic eruptions apparently cause a redistribution of infrasound along the TCs track which can lead to the formation of new TCs.
