Specific pathogen-free guinea pigs were infected via the respiratory route with viable, attenuated Mycobacterium tuberculosis H37Ra and maintained on purified isocaloric diets. The control diet contained 30% protein (ovalbumin) and 50 ppm of added zinc (50 micrograms/g), the low protein diet contained 10% protein and 50 ppm of added zinc, and the low zinc diet contained 30% protein and no added zinc. Guinea pigs from each diet treatment were skin tested with purified protein derivative 48 h before sacrifice at 3, 4, and 5 weeks postinfection. Protein-deficient animals exhibited significantly reduced body weight, spleen weight, serum total proteins, and serum albumin. Zinc deficiency was characterized by loss of weight and progressive reductions in plasma zinc concentrations. The number of viable M. tuberculosis H37Ra cells was significantly higher in the lungs of both malnourished groups at 3 weeks, but fell below control viable counts by 5 weeks postinfection. A similar pattern was seen in the spleens and bronchotracheal lymph nodes. Both the proportion and intensity of delayed hypersensitivity reactions increased steadily between 3 and 5 weeks in control animals, whereas the two malnourished groups were essentially anergic at all intervals, despite systemic infection. These results demonstrate that both protein and zinc deficiencies exert a significant influence on the development of pulmonary tuberculosis but that the nature of the influence depends upon the interval studied. In both malnourished groups, the pulmonary infection tended to peak early and decline, whereas the disease developed more slowly in control animals. Apparent control of mycobacterial populations in the tissues was accomplished by malnourished animals in the absence of demonstrable delayed hypersensitivity.

1. Zinc deficiency was produced experimentally in guinea-pigs fed on a diet containing 1.25 mg Zn/kg diet over a period of 60 d. In addition, the response of the Zn-deficient (ZnD) animals to Zn supplementation was studied for 15 d.2. In the ZnD group a significant reduction was found in serum Zn and protein concentrations and in alkaline phosphatase (EC 3. 1. 3. 1; AP) activity from day 24 onwards.3. Paper electrophoretic studies on serum revealed a significant decrease in relative values, as well as absolute values, of albumin and γ-globulin and an increase in β-globulin.4. Albumin:globulin increased on day 24 but decreased significantly from day 48 onwards.5. The kidney and testis of the ZnD group showed a reduction in Zn and protein contents, and AP activity.6. Zn supplementation of the previously ZnD group resulted in marked although incomplete improvement in the biochemical indices studied.

Helicobacter pylori Stores Nickel To Aid Its Host Colonization

The transition metal nickel (Ni) is critical for the pathogenicity of Helicobacter pylori. Indeed the element is a required component of two enzymes, hydrogenase and urease, that have been shown to be important for in vivo colonization of the host gastric mucosa. Urease accounts for up to 10% of the total cellular H. pylori protein content, and therefore the bacterial Ni demand is very high. H. pylori possess two small and abundant histidine-rich, Ni-binding proteins, Hpn and Hpn-like, whose physiological role in the host have not been investigated. In this study, special husbandry conditions were used to control Ni levels in the host (mouse), including the use of Ni-free versus Ni-supplemented food. The efficacy of each diet was confirmed by measuring the Ni concentrations in sera of mice fed with either diet. Colonization levels (based on rank tests) of the ⌬hpn ⌬hpn-like double mutants isolated from the mice provided Ni-deficient chow were statistically lower than those for mice given Ni in their diet. In contrast, H. pylori wild-type colonization levels were similar in both host groups (e.g., regardless of Ni levels). Our results indicate that the gastric pathogen H. pylori can utilize stored Ni via defined histidine-rich proteins to aid colonization of the host.
