Zinc finger proteins in cancer progression

Abstract: Zinc finger proteins are the largest transcription factor family in human genome. The diverse combinations and functions of zinc finger motifs make zinc finger proteins versatile in biological processes, including development, differentiation, metabolism and autophagy. Over the last few decades, increasing evidence reveals the potential roles of zinc finger proteins in cancer progression. However, the underlying mechanisms of zinc finger proteins in cancer progression vary in different cancer types and even in… Show more

“…Cellular zinc homeostasis is controlled by two families of zinc transporters with opposing actions, the ZnT family and the ZIP family . Dysregulation of zinc‐finger transcription factors and zinc transporters has been implicated in the progression of cancer . Specific to HCC, hepatoma cells have significantly lower intracellular zinc levels than normal hepatic cells, and the ZIP14 zinc transporter is markedly downregulated in the hepatoma cells .…”

“…The liver is responsible for production of carrier proteins for copper and zinc and is also the main site for copper storage, which plays a central role in maintaining copper and zinc homeostasis . A growing body of evidence has established a strong connection between the disruption of copper and/or zinc homeostasis and the development and progression of cancer . Data from case–control studies indicate higher copper levels, lower zinc levels, or higher copper/zinc ratio in patients with various cancers, including HCC .…”

“…GFI1 (Growth Factor Independent 1 Transcriptional Repressor) (4 sites) is a protein-coding gene, and its methylation was found to be the most robust mediator of the association between maternal smoking and birthweight in a recent study of children and newborns exposed to maternal and paternal smoking [47]. ZNF668 (2 sites) is a zinc finger protein whose role in DNA repair, cell proliferation and cancer has been investigated [48]. Chromosome region 1p36 (2 sites) has been investigated as a region containing a possible tumor suppressor [49]; with investigations including the effects of methylation [50].…”

“…Studies have shown that ZFX functions as a transcription factor [65] with the capacity to regulate self-renewal and cell survival in embryonic and haematopoietic stem cells [66,67]. ZFX has been recognised as an oncogene in a number of cancers (reviewed in [68]). In prostate cancer cell lines, ZFX has been shown to drive cell proliferation and promote cell survival [69] and expression of ZFX has been shown to be elevated in prostate tumours [69,70].…”
