Zinc- and bicarbonate-dependent ZIP8 transporter mediates selenite uptake

Abstract: Selenite (HSeO 3− ) is a monovalent anion of the essential trace element and micronutrient selenium (Se). In therapeutic concentrations, HSeO 3 − has been studied for treating certain cancers, serious inflammatory disorders, and septic shock. Little is known, however, about HSeO 3 − uptake into mammalian cells; until now, no mammalian HSeO 3 − uptake transporter has been identified.

“…Iron and zinc inhibit the ZIP8-mediated uptake of one another [40, 42, 43]. Therefore, these cations have the potential to modulate the impact of zinc on macrophage immune function through competitive uptake and cell signaling [44, 45].…”

“…Zrt/Irt-like protein 8 (ZIP8) is a multifunctional membrane transporter protein that facilitates uptake of several essential and nonessential divalent metals, including Zn 2ϩ , Mn 2ϩ , and Fe 2ϩ (15,18,34,57). More recently, ZIP8 was found to be a major transporter of inorganic selenite (HSeO 3 Ϫ ); transport of selenite requires both Zn 2ϩ and bicarbonate as cosubstrates (41). ZIP8 substrates Zn 2ϩ , Mn 2ϩ , Fe 2ϩ , and selenite are critically required in maintaining normal cellular function.…”

“…Cation transport by ZIP8 is pH dependent and electroneutral, indicating that it facilitates the co-transport of other ionic species as well [ 47 , 69 , 71 ]. ZIP8 participates in the cytosolic influx of manganese, cadmium, iron, zinc, and selenite [ 71 , 77 ]. Iron and zinc inhibit the ZIP8-mediated uptake of each other [ 69 ].…”
