Zika Virus Infection of Human Iris Pigment Epithelial Cells

Abstract: During recent Zika epidemics, adults infected with Zika virus (ZIKV) have developed organ-specific inflammatory complications. The most serious Zika-associated inflammatory eye disease is uveitis, which is commonly anterior in type, affecting both eyes and responding to corticosteroid eye drops. Mechanisms of Zika-associated anterior uveitis are unknown, but ZIKV has been identified in the aqueous humor of affected individuals. The iris pigment epithelium is a target cell population in viral anterior uveitis, … Show more

“…ZIKV-infected blood aqueous barrier (BAB) cells induce inflammation that damages the BAB and facilitate influx of effector immune cells into the anterior uvea. Influx of ZIKV-infected monocytes acting as Trojan horses [137][138][139].…”

“…ZIKV-infected corneal epithelial cells express TLR3, RIG-I, and MDA5 to facilitate the generation of type I and III interferons that mediate antiviral response [133,137]. Singh and coworkers [133] demonstrated that ZIKV-infected corneal epithelial cells support ZIKV replication as well as promote ZIKV persistence in the cornea.…”

“…The expression of IRF2 including IRF1 binding site at promoter region is inducible by transient or stable IRF1 expression [ 32 34 ]. IRF2 was up-regulated in ZIKV infection of human iris pigment epithelial cells [ 35 ]. In another study, IRF2 expression was induced during acute and latent infection [ 36 ].…”

“…The ARPE-19 human retinal epithelial cell line exhibits differences from primary human cells, including in the setting of infection [30]. Also, there are differences in gene expression between individuals that may be considerable [30,36,37]. Therefore, we sought to confirm the key finding of a difference in infectivity and host response between DENV-1 strains from the 2005 versus the 2007 Singapore epidemics using retinal pigment epithelial cells individually isolated from eyes of five different human donors.…”
