Zebrafish: An In Vivo Model for Nano EHS Studies

Abstract: To assure a responsible and sustainable growth of nanotechnology, the environmental health and safety (EHS) aspect of engineered nanomaterials and nano-related products needs to be addressed at a rate commensurate with the expansion of nanotechnology. Zebrafish has been demonstrated as a correlative in vivo vertebrate model for such task, and the current advances of using zebrafish for nano EHS studies are summarized here. In addition to morphological and histopathological observations, the accessibility of ge… Show more

“…To date, various animal models are in principle available to investigate and dissect the relevance of the NPs' biomolecule corona for human health. These are ranging from rodents to fish and other organisms, and have been reviewed before (17,19,31,(131)(132)(133)(134)(135). Notably, recent developments also applied successfully HTS/HCS approaches for whole animals, such as zebrafish (121,136).…”

“…Furthermore, the body of knowledge for ZF is small compared to that of rodents in terms of metabolism and pharmacokinetics. 85 To our knowledge, no comparative data exists for establishing when is it necessary to induce human tumors on an inherently angiogenic model in order to study angiogenesis. Researchers must determine what is necessary to do this by establishing a correlation between data in these two scenarios.…”

“…The NP or sample application step can present a bottleneck, especially if microangiography and injection is the exposure method of choice. 85 Automated high-throughput injection techniques are being developed 86 87 but have thus far only been applied to early embryo injection 87 and for injecting into non-vascular areas of ZF larvae. 86 However, the injection location can be altered by designing a specific macro for the software control program, 86 thus vascular injections will likely be tested soon.…”

“…The ZF is considered the gold standard for evaluating environmental toxicity (Scholz et al, 2008). With the expansion of nanotechnology, the ZF has been gaining ground as an experimental model for environmental health and safety tests of various nanomaterial (Lin et al, 2013;Scholz et al, 2008). The ZF model has been used for assessing the toxicity of silver nanoparticles (Muth-Köhne et al, 2013), silica (Duan et al, 2013) and other nanomaterials (Jevgenij et al, 2013).…”

“…This fact promotes and increases the relevance of their use in biological assays, which is hard to be matched in vitro (Lin et al, 2013). The objective of this work is to update knowledge on the use of ZF as an experimental model for biological assays with natural products and derived.…”
