Yusho patients show increased serum IL-17, IL-23, IL-1β, and TNFα levels more than 40 years after accidental polychlorinated biphenyl poisoning

Abstract: The Yusho poisoning incident, caused by rice oil contaminated with polychlorinated biphenyls (PCBs), polychlorinated quarterphenyls (PCQs), and polychlorinated dibenzofurans (PCDFs) generated by heat-denatured PCBs, occurred in 1968 in western Japan. Although severe symptoms are rarely observed today, the levels of PCBs and PCDFs in the sera of Yusho patients remain high. The aryl hydrocarbon receptor (AhR), which also acts as a dioxin receptor, is a transcriptional regulator that mediates dioxin toxicity. Rec… Show more

“…Conversely, IL-22 can have a protective role in acute inflammation state as arise during hepatitis and inflammatory bowel disease (Zenewicz et al 2007(Zenewicz et al , 2008. Reduced IL-22 expression level in dioxin-exposed people was consistent with results showing reduced IL-22 levels in PCB-contaminated Yusho patients (Kuwatsuka et al 2014). As various cell types can produce pro-inflammatory cytokines, further in vitro studies could reveal which types of immune system cells might be affecting the changes in inflammatory cytokine expression being noted in dioxin-exposed people.…”

“…POPs have been associated with increased inflammation, assessed by a variety of biomarkers (Fujiyoshi et al 2006;Imbeault et al 2012;Kumar et al 2014b;Kuwatsuka et al 2014). In the current study, we did not find any associations of CRP with POPs, and other investigators have inconsistently detected effects of POPs on CRP.…”

“…Another study, on the impact of POPs on human adipose cells, showed that both precursor cells and adipocytes are targets of POPs and that these pollutants trigger mainly the inflammation pathway (Kim MJ et al 2012). In a study from Japan involving 40 Yusho patients and 40 controls, Kawatsuka et al (2013) demonstrated that serum levels of interleukin (IL)-17, IL-1β, IL-23, and tumor necrosis factor-α (TNF-α) were higher in patients who were exposed to POPs, including PCBs through consumption of contaminated rice (Kuwatsuka et al 2013). Circulating inflammatory biomarkers such as CRP, IL-6, TNF-α, monocyte chemotactic protein 1 (MCP-1), intercellular adhesion molecule 1 (ICAM-1), vascular cell adhesion protein 1 (VCAM-1), and E-selectin have been associated with a variety of metabolic disorders and associated outcomes (Goldberg 2009).…”
