Hybrid adoption, irrigation, and planting density are important factors for maize (Zea mays L.) production in semiarid regions. For this study, a 2-yr field experiment was conducted in the Texas High Plains to investigate maize yield determination, seasonal evapotranspiration (ETc), and water-use efficiency (WUE) under limited irrigation. Two hybrids (N74R, a conventional hybrid, and N75H, a drought-tolerant (DT) hybrid) were planted at three water regimes (I 100 , I 75 , and I 50 , referring to 100%, 75%, and 50% of the evapotranspiration requirement) and three planting densities (PD 6, PD 8, and PD 10, referring to 6, 8, and 10 seeds m −2 ). At I 50 , drought stress reduced grain yield by 4.78 t/ha for the conventional hybrid but only 4.22 t/ha for the DT hybrid, when compared to I 100 . Although ETc decreased at I 75 and I 50 , the highest WUE was found at I 75 . The DT hybrid did not yield more than the conventional hybrid but had greater yield stability at lower water regimes and extracted less soil water. Drought decreased biomass, harvest index, and kernel weight but did not affect kernel number. Higher planting densities increased biomass and kernel number but decreased kernel weight. Kernel number and kernel weight of the conventional hybrid were more sensitive to planting density than the DT hybrid. These data demonstrated that limited irrigation at I 75 is an effective way to save water and maintain the maize yield in semiarid areas, and that DT hybrid shows a greater yield stability to plant density under water stress. 2-(3,4-Dichlorophenoxy) triethylamine (DCPTA) regulates plant development; however, the molecular basis of this regulation is poorly understood. In this study, RNA sequencing (RNA-seq) analysis and physiological indexes of maize seedlings (three-leaf stage) treated with 15% polyethylene glycol (PEG) with/without DCPTA were investigated to explore the possible mechanism of exogenous DCPTA-improved drought tolerance. In the library pair comparisons of DCPTA vs. the control, PEG vs. the control, and PEG + DCPTA vs. PEG, totals of 19, 38 and 20 differentially expressed genes (DEGs) were classified as being involved in metabolic processes, respectively; totals of 5, 11, and 6 DEGs were enriched in the nitrogen (N) metabolic pathway, respectively. The genes encoding nicotinamide adenine dinucleotide-nitrate reductase (NADH-NR), ferredoxin-nitrite reductase (Fd-NiR), reduced ferredoxin-glutamate synthase (Fd-GOGAT), and chloroplastic glutamine synthetase (GS 2) were common in response to PEG-simulated drought stress with/without DCPTA treatment. Moreover, DCPTA maintained stable gene relative expression levels and protein abundances of NADH-NR, Fd-NiR, GS2, and Fd-GOGAT. Moreover, exogenous DCPTA partially mitigated PEG-simulated drought-induced reductions in the enzymatic activities of NR, nitrite reductase (NiR), glutamine synthase (GS), glutamine oxoglutarate aminotransferase (GOGAT), and transaminase, as well as in the contents of nitrate (NO 3 − ), nitrite (NO 2 − ) and soluble proteins and increases in the contents of ammonium (NH 4 + ) and free amino acids. Together, our results indicate that exogenous DCPTA improved plant growth and drought tolerance by regulating N-mechanism enzymatic activities involved in transcription and enzymatic protein synthesis.

Response of maize hybrids under limited irrigation capacities: Crop water use

Maize (Zea mays L.) production in the semiarid Texas High Plains (THP) often suffers exceptional yield reductions when irrigation cannot meet seasonal and peak water requirements. Drought-tolerant hybrids may reduce yield losses resulting from water stress by altering seasonal soil water use patterns to permit greater uptake during yield-sensitive growth stages. We examined the effect of irrigation capacity (8.47 and 4.23 mm d −1 ) and planting rates (74,000 and 95,000 seeds ha −1 ) during three growing seasons (2016-2018) on water use of three maize hybrids, two of which were considered drought tolerant. Changes in stored soil water were examined for the entire profile and for segregated depth increments (0-0.4, 0.4-0.8, and 0.8-1.4 m).Seasonal crop evapotranspiration averaged 754 and 592 mm across years for the high and low irrigation capacities, respectively. Irrigation level significantly affected seasonal change in stored soil water in 1 yr (2018) with greater depletion at harvest under the low irrigation capacity. Soil water use at 0.8-1.4 m comprised <4% of seasonal water use. Hybrids did not differ with respect to growing season crop water use. Stored soil water throughout the growing season within the examined depth increments were similar among hybrids and plant densities, with significant differences occurring infrequently and being inconsistent across years. Drought-tolerant traits may have a diminished effect on transpiration in this study because of the dense soillimiting root proliferation or the high atmospheric demand at the study location.
