Yeast Microflora Isolated From Brazilian Cassava Roots: Taxonomical Classification Based on Molecular Identification

Abstract: A rapid molecular identification technique was applied on microbial microflora isolated from Brazilian cassava roots given a yeast profile presented in the samples analyzed. A total of 24 strain isolated from cassava were initially grouped and identified in five groups using restriction-fragment length polymorphism (RFLPs) of 5.8S-ITS rDNA region. Sequencing analysis of the domains D1 and D2 of the 26S rRNA gene or 5.8S rRNA-ITS region were used to identify different groups of yeasts. Representative colonies o… Show more

“…Finally, regarding Profile X, two of the total isolates were identified as R. nothofagi. In the case of yeasts included in Groups III, VIII and X, it would have been useful to introduce more restriction enzymes besides CfoI, HaeIII and HinfI to increase the discriminatory capacity of the ITS1-5.8S RNA-ITS2 PCR-RFLP method, as suggested in other works (Esteve-Zarzoso et al, 1999;Ferreira et al, 2010). This is especially important in the studying of the ecosystems that have not yet been researched.…”

“…Thirty blue colonies of each sample were randomly picked and the identity of isolates with the inoculant strain was confirmed by restriction fragment length polymorphism (RFLP) analysis of the 16S‐23S rRNA intergenic spacer region . For yeast detection, 30 colonies of each sample in one plate during total yeast enumeration were picked and the identity of isolates with the inoculant strain was confirmed by RFLP analysis of the 5.8S rRNA gene …”
