Y-box-binding protein 1 contributes to IL-7-mediated survival signaling in B-cell precursor acute lymphoblastic leukemia

Abstract: Abstract. Y-box-binding protein 1 (YB-1) is a regulatory protein that is associated with drug resistance and relapse in solid tumors. As YB-1 mediates some of its activity through growth factor receptor signaling dysregulation, the present study compared the expression of YB-1 and interleukin 7 (IL-7) receptor α (IL-7Rα) in pediatric B-cell precursor (BCP) acute lymphoblastic leukemia (ALL) and normal BCP cells. The expression levels of IL-7Rα and YB-1 were higher in relapsed vs. diagnostic samples of primary … Show more

“…Our results indicated that YBX-1 and IL-6 had significantly higher sensitivity and specificity than CA 15-3 which implies their potential role as prognostic markers for breast cancer. The prognostic role of YBX-1 and IL-6 has been referred to in a number of previous studies on various types of cancers including ovarian [36], prostate [37], acute lymphoblastic leukemia [38] and multiple myeloma [39]. However, most of these studies focused on analysis of either fresh or paraffin-imbedded tissues with few exceptions which focused on minimally invasive samples like serum or plasma [35] [40].…”

“…Additionally, IL-7 withdrawal blocks the activation of P38 and JNK kinases, leading to IL-7-dependent thymocyte death [ 71 ]. IL-7 rescues rapamycin-induced apoptosis of B-cell precursor acute lymphoblastic leukemia-acute lymphoblastic leukemia (ALL) cells by upregulating MEK/ERK [ 72 ]. Hence, the MAPK pathway may play a vital role in regulating cell development via IL-7-mediated signal transduction.…”
