Fusarium oxysporum f. sp. melonis produces extracellular endo-1,4-beta-xylanase and beta-xylosidase when grown in shaken culture at 26 degrees C in a mineral salts medium containing oat spelt xylan and glucose as carbon sources. Endo-1,4-beta-xylanase was purified 251 times from 5-day-old culture filtrates, by Sephacryl S-200, ion exchange and gel filtration HPLC. The purified sample yielded a single band in SDS polyacrylamide gels with a molecular mass of 80 kDa on electrophoretic mobility and 83 kDa by gel filtration behavior. High activity of the endo-1,4-beta-xylanase against xylan was observed between 5 and 8 pH, and between 40 and 60 degrees C, the optimum pH and temperature being 5.0 and 50 degrees C, respectively. Kinetic properties of the enzyme are similar to those of other fungal xylanases, showing high affinity towards oat spelt xylan with a Km of 1 mM expressed as xylose equivalent.

ABSTRAKEndo-β-1,4-D-xilanase merupakan enzim hidrolitik yang memotong ikatan 1,4 pada rantai polisakarida xilan. Telah dilakukan tranformasi plasmid pET-Endo berisikan gen penyandi endo-β-1,4-D-xilanase dari Bacillus sp. asal abdominal rayap kedalam E. coli BL21. E. coli BL21 yang mengandung plasmid pET-Endo dapat memproduksi enzim xilanase dalam jumlah besar atau skala produksi. Untuk menurunkan biaya produksi perlu dicari media alternatif pertumbuhan E. coli menggantikan media cair Luria Bertani (LB) yang harganya mahal. Eksplorasi dilakukan pada kaldu usus ayam potong yang harganya murah tapi mengandung banyak protein. Kandungan N terlarut kaldu usus ayam mencapai 87 % dari LB Cair. Pertumbuhan E. coli diamati berdasarkan nilai Optical Density (OD) dengan spektrofotometer. Variasi dilakukan pada suhu inkubasi dan konsentrasi penambahan glukosa pada kaldu usus ayam. Hasil penelitian menunjukkan bahwa pertumbuhan optimal E. coli BL21 pET-Endo pada penambahan glukosa 1,5 % dengan suhu inkubasi 37 o C selama 16 jam. Kondisi optimum ini digunakan untuk menumbuhkan E. coli BL21 pET-Endo guna memproduksi enzim xilanase. Purifikasi enzim dilakukan dengan kromatografi afinitas Ni-NTA. Pada elusi imidazol 100 mM diperoleh kadar protein terbesar yaitu 0,076 mg/mL. Karakteristik enzim xilanase dengan aktivitas sebesar 0,042 U/mL dan aktivitas spesifik sebesar 0,556 U/μg. Faktor kemurnian sebesar 3,16 kali dan berat molekul enzim sebesar ± 30.000 Dalton.Kata kunci : Endo-β-1,4-D-xilanase, E. Coli BL21, glukosa, kaldu usus ayam.
