Xanthones from the Bark of Garcinia xanthochymus and the Mechanism of Induced Apoptosis in Human Hepatocellular Carcinoma HepG2 Cells via the Mitochondrial Pathway

Abstract: Xanthones are important chemical constituents of Garcinia xanthochymus and varied bioactivities including cytotoxicity. However, their anti-tumor mechanism has remained unknown. Here, we isolated and identified a new xanthone named garciniaxanthone I (1) and five known compounds from the bark of G. xanthochymus. Their structures were elucidated by NMR analysis and HRESIMS. The anti-proliferation activities of all isolated compounds were evaluated on four human tumor cell lines (HepG2, A549, SGC7901, MCF-7). Th… Show more

“…In our previous studies, series of PPAPs and xanthones isolated from the barks and fruits of G. xanthochymus exhibited good antitumor activities [ 20 , 21 , 22 ]. To explore more functional chemical constituents as novel tools for cancer therapy, we isolated and identified six new PPAPs together with nine known compounds from the edible fruits of G. xanthochymus .…”

“…Caspase-8, a caspase protein encoded by the Casp8 gene, plays an essential role in cell apoptosis induced by death receptors including the TNF receptors, Fas/Apo1, and DR3 (Varfolomeev et al, 1998). Jin, Shi, Liu, Chen, and Yang (2019) . Therefore, caspase-8 exerts both prodeath and prosurvival effects in different circumstances.…”

“…Caspase‐8, a caspase protein encoded by the Casp8 gene, plays an essential role in cell apoptosis induced by death receptors including the TNF receptors, Fas/Apo1, and DR3 (Varfolomeev et al, 1998). Jin, Shi, Liu, Chen, and Yang (2019) showed that a new type of xanthones promoted HepG2 cell apoptosis through enhancing the expression of cleaved caspase‐8, caspase‐9, and caspase‐3. Qiu et al (2019) also found that apigenin inhibited cell proliferation and induced cell apoptosis by activating the caspase‐8 expression.…”
