Xanthomonas campestris pv. vesicatoria Secretes Proteases and Xylanases via the Xps Type II Secretion System and Outer Membrane Vesicles

Abstract: Many plant-pathogenic bacteria utilize type II secretion (T2S) systems to secrete degradative enzymes into the extracellular milieu. T2S substrates presumably mediate the degradation of plant cell wall components during the host-pathogen interaction and thus promote bacterial virulence. Previously, the Xps-T2S system from Xanthomonas campestris pv. vesicatoria was shown to contribute to extracellular protease activity and the secretion of a virulence-associated xylanase. The identities and functions of additio… Show more

“…On evolutionary time scales, public goods provided by OMVs may select for cooperative behavior, driving the establishment of mutualisms. Finally, we note that the preponderance of research has examined OMVs in the context of animal symbioses; to date, only a few studies have focused on OMV-plant interactions (23,(99)(100)(101)(102)(103). Extending the role of OMVs in the biology of other multicellular lineages is a budding area for future investigation.…”

“…Horseradish peroxidase-labeled anti-rabbit or anti-mouse antibodies (GE Healthcare) were used as secondary antibodies. For the analysis of protein secretion via outer membrane vesicles, bacteria were incubated on a rotary shaker overnight at 30°C and supernatants were incubated with proteinase K as described previously (44). Experiments were performed three times with similar results.…”

“…For the analysis of OMVmediated protein secretion, strain 85-10hpaHoof containing HpaH-c-Myc was cultivated in nutrient-yeast-glycerol (NYG) medium, and bacterial culture supernatants were incubated in the absence or presence of proteinase K. OMV cargo proteins are protected from degradation by proteinase K, as is observed for the T2S substrate XCV4360 -c-Myc (Fig. 5B) (44). However, this was not the case for HpaH-c-Myc, suggesting that HpaH is secreted independently of OMVs by an alternative transport pathway.…”
