X-ray induced rearrangements between germ-line limited and soma chromosomes of Acricotopus lucidus (Diptera, Chironomidae)

Abstract: The germ-line limited chromosomes (Ks) [K being derived from 'Keimbahn' (Bauer, 1970)] of Acricotopus lucidus were studied in gonial and differential mitosis. After C-banding the soma chromosomes (Ss) are stained only at their centromeric regions whereas the Ks exhibit centromeric, intercalary and terminal heterochromatin. By X-raying sperms it was attempted to transfer K sections on or into Ss in order to bring finally S-linked K sections to polytenization in the salivary glands, and to obtain more knowledge … Show more

“…These results strongly support our earlier hypothesis (Staiber and Thudium 1986), that during evolution Ks have developed from the Ss by rearrangements and by the formation and accumulation of repetitive elements ± germ line-specific repetitive sequences as demonstrated in this investigation.…”

“…A permanent preparation of polytene salivary gland chromosomes of an X-ray induced reciprocal translocation between soma chromosome SIII and a germ line limited chromosome (K) of A. lucidus made in 1980 (Staiber and Thudium, 1986) was used in this study for obtaining K chromatin by microdissection. The K section is translocated to the long arm of SIII (III/6) ( Fig.…”

“…A permanent preparation of polytene salivary gland chromosomes of Acricotopus lucidus (Diptera, Chironomidae) from April 1980 carrying an X-ray induced rearrangment between a germ line-limited chromosome and a somatic one was used for microdissection (Staiber and Thudium, 1986). For chromosome preparation larval salivary glands had been fixed in ethanol:glacial acetic acid (3:1, v/v; 2 h), stained with acetic-carmine (12 h) and aceticlactic orcein (1.5 h) and squashed in a drop of the latter.…”

“…Some authors see the Ks as descendants of the S complement by way of endopolyploidy (Nicklas 1960;Painter 1966;Kunz et al 1970). First evidence for this hypothesis came from the analysis of X-ray-induced K-S rearrangements in polytene salivary gland chromosomes of A. lucidus, demonstrating that the Ks contain sections with S-homologous banding patterns (Staiber and Thudium 1986;Staiber 1991b).…”
