Total hydroxyproline in urine and free hydroxyproline, free and peptide hydroxyproline and protein bound hydroxyproline in serum are measured during wound healing in rats. The free and free+peptide fractions vary in concert with each other and with urine hydroxyproline. Protein bound hydroxyproline fractions take a very different course and behave as an acute phase reactant. The results suggest that protein bound hydroxyproline does not mirror collagen turnover but may be more relevant to C1q or complement metabolism.

Zusammenfassung Die vorliegende Arbeit befaßt sich mit der routinemäßigen Bestimmung des freien Hydroxyprolins im Serum von Pferden. Neben der einschlägigen Literatur wird die dabei angewandte Methodik und ihre Eignung für ein Routinelabor diskutiert. Außerdem wurden Normalwerte vom Pferde ermittelt, Tiere auf tageszeitliche Schwankungen von Serumwerten hin untersucht sowie die Stabilität des freien Hydroxyprolins unter verschiedenen Lagerungsbedingungen geprüft. Die beschriebene Methode liefert genaue, reproduzierbare Werte. Die FHP‐Serumkonzentrationen korrelieren mit dem Alter des Pferdes. Sie betragen danach bei zweijährigen Pferden im Durchschnitt 5,14 μg/ml, dreijährigen 4,79 μg/ml, vierjährigen 3,27 μg/ml, fünfjährigen 2,45 μg/ml, sechs‐ bis vierzehnjährigen 2,02 μg/ml und bei über fünfzehn Jahre alten Tieren 1,51 μg/ml. Einzelwerte von Fohlen liegen nach unserer Untersuchung je nach Alter zwischen 9,6 und 27,5 μg/ml. Das freie Hydroxyprolin unterliegt im Blut keinen tageszeitlichen Schwankungen und ist bei Aufbewahrung bei +4°C bis zu 5 Tagen sowie bei −20°C mindestens 3 Wochen sicher stabil. Summary The routine determination of free hydroxyproline in horse serum Methods and normal values The literature relating to the various methods for determining free hydroxyproline (FHP) is reviewed and the suitability of the methods for routine use is discussed. Normal values for the horse are given, and also time‐of‐day variations and the stability of free hydroxyproline under different storage conditions. The method described provides precise, reproducible results. The FHP serum levels are correlated with age of the animal. In 2‐year old horses they average 5.14 μg./ml., 3‐year old horses 4.79, 4‐year olds 4.27, 5‐year olds 2.45, 6–14‐year olds 2.02 and in horses over 15 years old 1.51 μg./ml. Individual values for foals, depending on their age, lie between 9.6 and 27.5 μg./ml. FHP shows no variations in blood depending on time of day and when stored at + 4 deg. C for up to 5 days or −20 deg. C for at least 3 weeks it remains stable. Résumé Détermination de routine de l'hydroxyproline libre dans le sérum de cheval Méthode et valeurs normales Le travail présent traite de la détermination de routine de l'hydroxyproline dans le sérum de chevaux. En plus des ouvrages se rapportant au sujet, on discute la méthode employee et son aptitude à un travail de routine en laboratoire. On donne les valeurs normales chez des chevaux en ayant examiné les variations journalières des valeurs sériques et la stabilité de l'hydroxyproline les variations journalièrs des valeurs sériques et la stabilité de l'hydroxyproline en fonction de conditions différentes. Les méthodes décrites donnent des valeurs exactes et reproductibles. Les concentrations sériques en FHP correspondent à l'âge du cheval. Les résultats chez les chevaux sont les suivants: 5,14 μg/ml à 2 ans, 4,79 μg/ml à 3 ans, 3,27μg/ml à 4 ans, 2,45 μg/ml à 5 ans, 2,02μg/ml de 6–14 ans et 1,51 μg/ml au‐dessus de 15 ans.
