Worldwide increased prevalence of human adenovirus type 3 (HAdV-3) respiratory infections is well correlated with heterogeneous hypervariable regions (HVRs) of hexon

Abstract: Human adenovirus type 3 (HAdV-3) respiratory infections occurs worldwide in both children and adults, leading to severe morbidity and mortality, particularly in the paediatric age group and especially in neonates. During HAdV infection, neutralizing antibodies are formed against the epitopes located in the hyper variable regions (HVRs) of the hexon protein. These neutralizing antibodies provide protection against reinfection by viruses of the same type. Therefore it is reasonable to speculate that variations o… Show more

“…These mutations contained the substitutions of hydrophilic/hydrophobic AA (T429A, A439D, T445A, and T159I) and insertion of heterocyclic AA (20P). Similar AA substitutions in hexon have been observed in strains circulating in Japan, Korea, Germany, and Taiwan, China (Haque et al, 2018). The locations of the mutations in our study were not included in the epitopes that have been functionally confirmed in previous reports (Leen et al, 2004;Chakupurakal et al, 2013;Keib et al, 2017).…”

“…Surprisingly, a recent case of HAdV-C2 caused meningoencephalitis was also reported in a middle-aged woman in the US 4 . HAdV have a high prevalence 58 and are broadly used as gene therapy vectors 9 and oncolytic viruses 10,11 . The high seroprevalence of HAdV-C2/5 12 underlines that HAdV infections are asymptomatic in healthy individuals, but HAdV persist in mucosal lymphocytes, and thereby pose a severe risk for immunosuppressed patients undergoing stem cell transplantation 13 .…”

“…They cause morbidity and mortality, especially to immuno-compromised patients 1,2 as indicated by a recent outbreak in the USA killing 12 children, or a recent case of meningoencephalitis in a middle-aged woman in the USA 3 . HAdVs have a high prevalence [4][5][6][7] and are broadly used as gene therapy and vaccination vectors as well as oncolytic viruses [8][9][10] . The high seroprevalence of HAdV-C2 and C5 (species C, types 2 and 5) underlines that HAdV infections are asymptomatic in healthy individuals, but persist in mucosal lymphocytes, and thereby pose a risk for immunosuppressed patients undergoing stem cell transplantation 11,12 .…”
