Within-day Physiologic Variation of Leukocyte Types in Healthy Subjects as Assayed by Two Automated Leukocyte Differential Analyzers

Abstract: The physiologic within-day (700-2200 h) variation of leukocyte-type concentrations in blood as determined for 21 healthy young adults is reported. All blood specimens were obtained in duplicate such that the within-batch analytic variation, as well as the pertinent biologic sources of variation, was able to be determined. All specimens were analyzed on each of two automated leukocyte differential systems: Hemalog-D Differential System and the Hematrak-240 Analyzer. On the basis of a comparison of the performan… Show more

“…The utility of hematologic biomarkers of glucocorticoid sensitivity might be substantially improved by intra-individual assessments over time (e.g., repeated measurements of cortisol and leukocyte subset distributions over their diurnal cycle (Winkel, Statland et al 1981)). That would hold constant other hematologic influences that likely added substantial noise to the present cross-sectional analysis (and likely explain why relationships between cortisol levels and hematologic parameters rarely exceeded r = .20 in magnitude).…”

“…1 Several previous studies have investigated the biological variations of hematologic parameters. [2][3][4][5][6][7][8][9] Some parameters, such as hemoglobin or reticulocytes, have been demonstrated to exhibit hour-to-hour, day-to-day, or seasonal intraindividual fluctuation. 3,6 We revisit these areas for several reasons.…”

“…Second, almost all the reported studies in the past have been based entirely on Caucasian/white populations. [2][3][4][5][6][7][8][9] There is little knowledge about biological variation of hematologic parameters among Asian populations. Third, many new hematologic parameters have become available from modern automated hematology analyzers in recent years.…”
