Wilms' tumor 1 (WT1) expression and prognosis in solid cancer patients: a systematic review and meta-analysis

Abstract: Though proposed as a promising target antigen for cancer immunotherapy, the prognostic value of Wilms' tumor 1 (WT1) in solid tumors remains inconclusive. Here, we report a systematic review and meta-analysis of the association between WT1 expression and prognosis in solid tumors. PubMed, Web of Science and Google Scholar were searched to identify studies exploring the impact of WT1 on clinical outcomes, including overall survival (OS), disease-specific survival (DSS), disease-free survival (DFS), relapse/recu… Show more

“…This enrichment indicates that WT1 has the ability to influence many of the properties of malignancy through transcriptional regulation of target genes, consistent with the well-documented role for WT1 in tumor development. 2,32,34 Another interesting group is nucleocytoplasmic transport. Accumulating evidence suggests that transport from the nucleus to cytoplasm is tightly regulated and at least in some instances selective for certain transcripts, 35 potentially providing WT1 with further possibilities to influence protein production.…”

“…WT4869 is a novel peptide derived from the tumor-associated antigen Wilms' tumor gene 1 (WT1), which is commonly overexpressed in leukemias, MDS (7) and a variety of solid tumors. (8) WT1 is ranked the most useful among 75 cancer antigens by the National Institutes of Health, and is attracting attention as a peptide suited for use as a cancer vaccine. (9) The WT1 235-243 peptide derived from the WT1 gene product is restricted to the human leukocyte antigen (HLA)-A*24:02 that is present in approximately 60% of the Japanese population.…”

“…The oncogenic roles during tumorigenesis, such as promotion of growth, inhibition of differentiation, resistance to cell death, and promotion of tumor angiogenesis (8)(9)(10)20). Further, wildtype WT1 is overexpressed in various human cancers including PDAC and is a poor prognostic marker (11)(12)(13)(14). In addition to these oncogenic functions, WT1 is immunogenic.…”
