Background: Despite the benefits of regular physical activity (PA), most adults with multiple sclerosis (MS) are insufficiently active. Identifying the motivational correlates of PA is necessary to facilitate health behavior change. The extent to which the constructs of psychological need satisfaction and motivational regulations associate with self-determined PA in adults with the disease was examined. Methods: Individuals with MS were provided a link to a web-based survey. There were 290 respondents: 242 women and 48 men aged 22 to 71 (mean ± SD, 49.50 ± 12.05) years with primarily mild-to-moderate mobility impairment who completed the Psychological Need Satisfaction in Exercise scale, the Behavioral Regulation in Exercise Questionnaire, and the International Physical Activity Questionnaire. Results: Path analysis revealed that PA was best predicted by integrated regulation, competence, and mobility, explaining 28% of the variance in PA behavior. All three need satisfaction variables (relatedness, competence, and autonomy) and mobility impairment accounted for 43% of the variance in integrated regulation. Conclusions: Increasing satisfaction of the need for relatedness, competence, and autonomy can lead to more integrated and internally motivated PA engagement in adults with MS.

Alternative splicing is a potent modifier of protein function. Stromal interaction molecule 1 (Stim1) is the essential activator molecule of store-operated Ca 2+ entry (SOCE) and a sorting regulator of certain ER proteins such as Stimulator of interferon genes (STING). Here, we characterize a conserved new variant, Stim1A, where splice-insertion translates into an additional C-terminal domain. We find prominent expression of exonA mRNA in testes, astrocytes, kidney and heart and confirm Stim1A protein in Western blot of testes. In situ, endogenous Stim1 with domain A, but not Stim1 without domain A localizes to unique adhesion junctions and to specialized membrane retrieval sites (tubulobulbar complexes) in testes. Functionally, using Ca 2+ imaging and patch-clamp analysis, Stim1A shows a dominant-negative effect on SOCE and I CRAC , despite normal clustering and interaction with Orai1 investigated by combined TIRF and FRET analyses and as confirmed by an increased SOCE upon knock-down of endogenous Stim1A in astrocytes. Mutational analyses in conjunction with imaging and patch-clamp analyses of residues either in domain A or within the N-terminal region of Orai1 demonstrate a specific defect in stabilized channel gating. РезюмеОбоснование: качество жизни (КЖ) считается интегральной характеристикой физического, психологического, эмоционального и социального функционирования больного. Улучшение качества жизни рассматривается как одна из задач терапевтического вмешательства при ведении больных рассеянным склерозом (РС). Цель: сравнительный анализ разных методов оценки качества жизни больных рассеянным склерозом с депрессивным расстройством. Пациенты и методы исследования: обследованы 203 больных рассеянным склерозом (56 мужчин и 147 женщин) в возрасте от 15 до 60 лет (средний возраст 39,7 ± 10,91). Диагноз РС устанавливался в соответствии с критериями McDonald 2010, 2017 гг. Среди больных РС оказалось 113 пациентов с депрессией. В группы сравнения вошли 90 больных РС без депрессии и 70 больных депрессией без неврологического заболевания. В группу контроля включены 52 здоровых добровольца в возрасте 29,25 ± 5,12 года. Для оценки КЖ больных использовали краткий опросник самооценки здоровья (The Short Form-36, SF-36) и опросник качества жизни больных рассеянным склерозом (Multiple Sclerosis Quality of Life-54, MSQOL-54). Результаты: сравнительная оценка результатов показала, что использование опросников SF-36 и MSQoL-54 помогает более полно оценить качество жизни больных с неврологическими заболеваниями и депрессией. Каждый из инструментов дополняет информацию о состоянии больных в зависимости от цели и нозологии. Предложенные опросники могут быть рекомендованы для динамического наблюдения изменений качества жизни больных РС, а также для применения в клинических исследованиях и при подборе лекарственной терапии.
