Whole-Transcriptome Analysis of Mouse Models with MPTP-Induced Early Stages of Parkinson’s Disease Reveals Stage-Specific Response of Transcriptome and a Possible Role of Myelin-Linked Genes in Neurodegeneration

Abstract: Parkinson's disease (PD) is characterized by degeneration of dopaminergic neurons. A whole-transcriptome analysis of the substantia nigra and striatum of an MPTP-induced mouse models of the earliest stages of PD was performed. Functional clustering of differentially represented transcripts revealed processes associated with the functioning of synapses, dendrites, axons, and myelination of neuronal projections. All of these processes occur in both the substantia nigra and striatum, but they are aimed at the fun… Show more

“…7b). studies mainly focus on the transcriptome analysis of ST and substantia nigra [16], but less on HP, CC and CB regions. Emerging data suggest interactions between the dopaminergic systems and the hippocampus in synaptic plasticity, adaptive memory, and motivated behaviour.…”

“… 2 The clinical manifestations of PD include rigidity, posture instability, bradykinesia, and resting tremors. 3 Non‐motor symptoms include dysosmia, anxiety, and memory disorders. 4 Currently, the main treatment used for PD includes levodopa‐based drug therapy and deep brain stimulation surgery.…”

“…Brain samples were dissected to isolate the CC, HP, ST, and CB regions. Previous studies have primarily focused on the transcriptome analysis of the ST and substantia nigra brain regions [56], but few have studied the HP, CC, and CB regions. Emerging data suggest that there are interactions between the dopaminergic system and the HP, and these are involved in synaptic plasticity, adaptive memory, and motivated behavior.…”
