Whole organ vascular casting and microCT examination of the human placental vascular tree reveals novel alterations associated with pregnancy disease

Abstract: Experimental methods that allow examination of the intact vascular network of large organs, such as the human placenta are limited, preventing adequate comparison of normal and abnormal vascular development in pregnancy disease. Our aims were (i) to devise an effective technique for three-dimensional analyses of human placental vessels; (ii) demonstrate the utility of the technique in the comparison of placental vessel networks in normal and fetal growth restriction (FGR) complicated pregnancies. Radiopaque pl… Show more

“…Imaging of the fetoplacental vascular system can be enhanced by perfusing contrast agents into the fetal circulation to image the arterial and venous circulation (Junaid et al . ). From this, information on vessel branching patterns, interbranching length and capillary loop dilatations are useful in predicting the placenta's ability to optimally transfer oxygen, a portion of the placenta often inaccessible by other means (Junaid et al .…”

“…From this, information on vessel branching patterns, interbranching length and capillary loop dilatations are useful in predicting the placenta's ability to optimally transfer oxygen, a portion of the placenta often inaccessible by other means (Junaid et al . ). However, while microCT can image large regions of tissue, when doing this its ability to image the microcirculation is limited.…”

“…This provides a negative replica of the hollow space filled with the resin, which can be visualised after chemical maceration (corrosion) of the soft tissue. The combination of microvascular corrosion casting with scanning electron microscopy and micro-CT enables the three-dimensional analysis of the vasculature at high resolution (Aharinejad & Lametschwandtner, 1992;De Spiegelaere et al, 2010;Debbaut et al, 2014;Junaid, Bradley, Lewis, Aplin, & Johnstone, 2017;Murakami, 1971).…”
