Whole‐genome copy number and immunohistochemical analyses on surgically resected intracholecystic papillary neoplasms

Abstract: Intracholecystic papillary neoplasms are newly defined precancerous lesions. According to Classification of the World Health Organization, they have four histological morphologies, which are biliary, gastric, intestinal, and oncocytic. This study evaluated 17 patients with resected intracholecystic papillary neoplasms in terms of histological, immunohistochemical, and copy number variation (CNV). The histological subtypes included 5 cases of low‐grade (5 gastric) and 12 cases of high‐grade (6 gastric and 6 bil… Show more

“…The accumulation of genetic mutations with tumor progression was suggested in the previous case series of ICPN, and ICPN exhibited slight p53 and MIB-1 staining, reflecting its mutation in a previous case series [ 18 ]. Positive staining for β-catenin suggested activation of the Wnt/β-catenin pathway in ICPN [ 19 ].…”

“…Additionally, the risk-score formula integrated all genes, and two groups with different risk could be found among the results. Macrophages release interferon alpha-8 ( IFNA8 ) [ 26 ], and we found that the high-risk group was associated with high expression of CD163 , which is a marker of M2-like tumor-associated macrophages (TAMs). Caspase-1 ( CASP1 ) is involved in various inflammatory processes and initiates programmed cell death [ 31 ], and we found that the high-risk group also associated with high MYD88 expression.…”

“…The staining was performed using a Leica Bon-Max slide stainer (Leica Biosystems K.K. Tokyo, Japan) following the manufacturer’s instructions [ 26 ], Bond epitope retrieval solution 1 (pH 6.0, 30 min., #AR9961, Leica), and at a 1:400 dilution. After staining, the slides were scanned in a NanoZoomer S360 digital slide scanner (#C13220-01, Hamamatsu K.K.…”

“…The immunohistochemistry was performed using an automated piece of equipment, Leica BOND-MAX stainer, following the manufacturer’s instructions and as previously described [ 53 , 59 , 63 , 64 , 65 ]. The RGS1 primary antibody (rabbit polyclonal) was purchased from Thermofisher [ 63 ].…”
