WHIRLY1 recruits the histone deacetylase HDA15 repressing leaf senescence and flowering in Arabidopsis

Abstract: Leaf senescence is controlled by a complex regulatory network in which robustness is ensured by the activity of transcription factors and epigenetic regulators. However, how these coordinate the process of leaf senescence remains poorly understood. We found that WHIRLY1 interacts with Histone Deacetylase (HDA)15, a Reduced Potassium Dependence3 (RPD3)/ HDA1-type HDA, by using green fluorescent protein-nanotrap-mass spectrum assays. The development-dependent interaction between WHIRLY1 and HDA15 was further con… Show more

“…Our results indicate that WHIRLY1 may act as a scaffold protein in these complexes. WHIRLY1 physically interacts with histone deacetylase HDA15, as shown by yeast two-hybrid, bimolecular fluorescence complementation and co-immunoprecipitation, and both proteins co-localize to the region near the transcription start site of some nutrient-recycling-related genes [ 55 ]. This led to the assumption that WHIRLY1 indeed acts as a repressor via recruiting HDA15, which causes the deacetylation of associated histones.…”

“…The oeUPL3-GFP , oeUBP12-GFP , and oeGFP plants from the fifth to eighth rosette leaves of 6-wk-old plants were harvested and ground in liquid nitrogen. Total proteins were extracted from 1 g of materials and performed CoIP according to the description by Huang et al (2022) .…”

“…A study showed that C2H2-type zinc finger MdZAT10 can accelerate the aging of apple leaves ( Yang et al., 2021 ). In Arabidopsis, knocking out HDA15 can accelerate leaf aging and promote plant flowering ( Huang et al., 2022 ). In safflower, CtCP1 promotes the hydrolysis of the CtbHLH41 protein, accelerating the transcriptional activity of related genes and leaf senescence ( Hong et al., 2022 ).…”
