Wheat Storage Proteins in Transgenic Rice Endosperm

Abstract: Transgenic rice seed expressing wheat HMW glutenin subunit was characterized to study the effects of the wheat prolamin on the protein expression pattern and protein size distribution in the endosperm and the functional and rheological properties of the rice flour and dough. Significant differences were found in the protein expression pattern between the transgenic and wild type samples. Comparing the protein expression profiles of transgenic and nontransgenic plants, combined with proteomic-based studies, ind… Show more

“…The expression of wheat HMW-GSs, 1Dx5_KK, and 1Dy10_JK, was observed in KK and JK transgenic lines, respectively. Oszvald et al [ 32 , 33 ] reported that the significant over-expression of 1Dx5 in rice seeds results in changing storage protein composition and increasing disulfide isomerase (PDI) levels, causing a positive effect on the functional properties of rice dough. Previous studies reported that the significant suppression of one SSP is compensated for by the increased accumulation of other SSPs, including PDI [ 1 , 40 , 41 ].…”

“…In addition, the over-expression of wheat 1Dx5 and/or 1Dy10 HMW-GSs improves the extensibility and elasticity of dough from wheat [ 26 , 27 , 28 ] and rye [ 29 ]. In efforts that were aimed at improving rice dough quality, preliminary attempts have been made to express wheat SSPs, including 1Ax1 [ 30 ], 1Bx7 [ 31 ], 1Dx5 [ 32 , 33 ], and 1Dy10 [ 34 ] in rice seeds, although to date, only 1Dx5 has been shown to confer a positive effect on rice dough properties. The objective of this study was to engineer wheat HMW-GS into rice seeds.…”

“…Proteomic analysis of the transgenic line (Oszvald et al , 2013) showed that the levels of some molecular chaperones were significantly higher compared with non-transformed, control rice lines. This suggests that although the recombinant HMW-GS represents <0·8 % of the total storage proteins, its expression may be enough to trigger the ER stress response and therefore affect protein sorting and PB formation.…”

“…Studies on the GM modification of such subunits may also lead to the production of novel proteins encoded by altered versions of either the transforming or endogenous genes (Blechl and Vensel, 2013). A relevant similar study is that on transgenic rice seed expressing the wheat HMW subunit (Oszvald et al, 2013). Another aspect of this type of study that has importance in any future regulatory submission is the determination of potential changes in the allergenicity of the GM material (Lupi et al, 2013).…”
