What has single‐cell RNA sequencing revealed about microglial neuroimmunology?

Abstract: The use of single‐cell RNA sequencing (scRNA‐seq) in microglial research is increasing rapidly. The basic workflow of this approach consists of isolating single cells, followed by sequencing. scRNA‐seq is capable of examining microglial heterogeneity on a cellular level. However, the results gained from applying this technique suffer from discrepancies due to differences between applied methods characteristics such as the number of cells sequenced and the depth of sequencing. This review aims to shed more ligh… Show more

“…LFA1 also was shown to play an important role in Tregs CTLA4 mediated suppression as it is (does what?). A similar pathway to CTLA4 mediated suppression by Tregs vertebrates, is unlikely to occur in lampreys as it lacks CTAL4 as well as CD80, CD86 and CD28 as well as LFA1 (figure 1)[26]. We found that TGFB2 is expressed in lampreys along with GP130 and TRAIL.…”

“…This hypothesis is further supported by the observation that knocking out the lymphocytes Llfa1 gene which is responsible for the interaction with the endothelial cells during the first phase of transmigration resulted in deformations and changing the biomechanical aspects of Th1, including effects on their filopodia and lamellipodia, but not Th17 [ 69 ]. We postulate that single cell RNA-seq analysis could shed more light on CD4+ T cells heterogeneous interactions with the endothelial cells of the BBB [ 70 ]. To summarize, PECAM1 and CAV1 which are responsible for regulating the formation of the space for migration in transcellular and paracellular migration receptively are mutually exclusive mechanosensing and have different phylogenetic origin and history.…”

“…With single-cell ribonucleic acid (RNA) sequencing (scRNA-seq), several microglia phenotypes with complex functional potential have been defined in both healthy and disease states [ 35 , 36 , 37 ]. In a recent study, the unique molecular signature of the cells composing the SCI site in a mouse mid-thoracic contusion model was characterized with scRNA-seq.…”
