Volume expansion of erythrocytes is not the only mechanism responsible for the protection by arginine-based surfactants against hypotonic hemolysis

Abstract: Highlights Arginine-based surfactant Bz-Arg-NHC12 has both hemolytic and antihemolytic effects.  Both processes depend on the lipid composition of the erythrocyte membrane.  SRBC is more resistant than HRBC to the hemolytic effect of the surfactant.

“…This protective effect is often observed at surfactant concentrations below the corresponding CMC value. However, our previous published work demonstrated that Bz-Arg-NHC n display both hemolytic and protective activity above this parameter (Fait et al 2018). Moreover, Joondan and coworkers proved a similar trend for the hemolytic activity of aromatic amino acid-based surfactants, evidencing the crucial role of the surfactants' aggregates in the disruption of the membrane structure (Joondan et al 2016(Joondan et al , 2017Fait et al 2018).…”

“…Those authors reported that the QAS derivatives induced a signi cant cell-wall thickening as well as membrane deformations along with lipiddroplet formation in the cytoplasm, acting also as oxidative-stress stimulants. Bz-Arg-NHC n was previously found to induce both membrane permeabilization and induction of oxidative stress as part of their antifungal mechanisms (Fait et al 2018). The combination of these effects may also alter the cell wall, as evidenced in the cell-roughness and stiffness changes in the Bz-Arg-NHC n -treated yeasts.…”

“…However, the boundary between those two mechanisms is not clearly defined and may occur simultaneously. 45,47 It was assumed that if hemolysis took place through the osmotic hemolysis mechanism, then it could be protected by solutes with appropriate sizes. 46 In this work, to determine the hemolysis mechanism of PL-C n -x and the size of the resulting pores on the hRBC membrane, an osmotic protectant experiment was performed.…”
