Vocal learning-associated convergent evolution in mammalian proteins and regulatory elements

Abstract: Vocal learning, the ability to modify vocal behavior based on experience, is a convergently evolved trait in birds and mammals. To identify genomic elements associated with vocal learning, we integrated new experiments conducted in the brain of the Egyptian fruit bat with analyses of the genomes of 222 placental mammals. We first identified an anatomically specialized region of the bat motor cortex containing direct monosynaptic projections to laryngeal motoneurons. Using wireless neural recordings of this bra… Show more

“…The dashed lines show phylolm lines of best fit for each clade as a visual aid. (C) We also applied TACIT to associate motor cortex open chromatin with vocal learning, a binary trait, and here we show results for the Egyptian fruit bat open chromatin region PVIL01002568.1:139004-139596 in the GALC locus (permulations p FDR =0.0376), which was also associated with open chromatin specifically active in the orofacial motor cortex of bat 97 and which has previously been associated with severe speech and motor disabilities in humans 99 .…”

“…Zoonomia realizes that vision. In combination with existing resources such as ENCODE 38 and TOPMed 39 and new technologies including regulatory genomic assays 94, 97 , chromatin conformation capture 27, 51 , and massively parallel reporter assays 36 , Zoonomia’s alignments of hundreds of placental species address fundamental questions in mammalian evolution.…”

“…Stimulation of the vLMC 10,11 or dLMC 12 results in laryngeal contractions. Bats, which represent another clade of vocal learning mammals, have recently been shown to also possess these direct projections from M1 13,14 . Songbirds have analogous direct projections from the robust nucleus of the arcopallium (RA) to syringeal motor neurons in nucleus XII in the brainstem ( Figure 1B ) 15,16 .…”

“…Previous conclusions on vocal learning abilities are weakened by the lack of reliable identification of vocalizing individuals 911 . Combined, these gaps limit thorough exploration of the neural mechanisms of vocal production learning in this species, although cutting-edge technologies for measuring brain activity are readily available 1215…”
