Objective: The objective of the study was to investigate the protective effects of Vitex negundo (VN) extracts against ultraviolet (UV) light C-induced oxidative stress in human corneal epithelial cells (HCEC) in vitro. Methods: The VN extracts were checked for their antioxidant activities by biochemical tests. The preliminary phytochemical analysis was done to check the presence of secondary metabolites. To trigger oxidative stress, the HCEC were exposed to UVC irradiation (200 J/m2). Then, different concentration of VN extracts was administered after 1 h of UVC irradiation. The effects of UVC irradiation and the extracts on the HCEC viability, morphology, cell migration by scratch assay, and reactive oxygen species (ROS) production by dichlorofluorescein diacetate method were measured. Results: VN ethanol (VNE) extract showed the highest antioxidant activity than the aqueous extract. Phytochemical screening shows the presence of alkaloids, carbohydrates, proteins, phenolics, and tannins. The extracts showed no cytotoxicity on the HCEC and also the VNE extract did not alter the morphology of the cells. Results suggest that the VNE extract enhanced the proliferation and migration of HCEC. The VNE extract significantly decreased UVC-induced oxidative toxicity by inhibiting the intracellular ROS production and maintained cell viability. Conclusion: VN extracts with its strong antioxidant potential protects HCEC from UVC-induced oxidative stress. The results suggest that these extracts can be used as prophylactic agents or as a therapeutic agent in overcoming UV light-induced damage in various ocular surface diseases such as pterygium and photokeratitis.

Holy Basil—Ocimumtenuiflorum—is one of the popular new “superfoods” thought to act as an antioxidant and to reduce stress and anxiety. However, it is often surrogated with other Ocimum species differing in their chemical profiles that may even pose health risks to the consumers. Moreover, even specific chemotypes of Holy Basil itself can be toxicologically relevant, because they sometimes contain the carcinogen compound methyleugenol. Using DNA barcoding based on plastidic markers, O.tenuiflorum can be differentiated from other species of Ocimum. However, this approach is still suboptimal in handling larger sample numbers and in tracing chemotypes that accumulate methyleugenol. We have, therefore, designed a trait-related DNA barcode based on the enzyme eugenol O-methyltransferase (EOMT), responsible for the synthesis of methyleugenol. We show that a multiplex PCR combining trait-related and trait-independent markers can differentiate O.tenuiflorum from other Ocimum species and identify methyleugenol chemotypes of O.tenuiflorum, even in dried material sold as mixtures.

Therapeutic Potential of Traditional Indian Herbal Medicine in COVID-19: A Narrative Review

Background: Coronavirus disease 2019 (COVID-19) has become a global challenge to the health care system. A novel agent to combat this deadly virus is still a matter of research. Herbal molecules have served humanity since the beginning. Objectives: This narrative review aims to study the antiviral properties of medicinal plants, which are already effectively used in the past against various viruses. It derives the importance of exploration of such phytochemicals, which can be complementarily used to treat COVID-19. Methods: Studies related to traditional medicine and treatment for viruses were retrieved from databases including PubMed, Google scholar until December 2020 using the keywords SARS-CoV-2, COVID-19, Immunological, Phyto-chemicals, Traditional Medicine. The resulting publications were analyzed to develop a narrative review on the traditional Indian phytochemicals that have been shown to effectively treat various viral infections and potentially treat or prevent COVID-19. Results: Many of the researches are showing that Indian herbal compounds have a significant potential against viral diseases. Plants like Azadirachta indica, Withania somnifera, Tinospora cordifolia, Ocimum basilicum, and many more have been shown tremendous antiviral, anti-inflammatory, and immune-modulatory activities. Conclusion: Phytochemicals obtained from the herbs can be helpful in the treatment and prevention of SARS-CoV-2via various modes such as inhibition of attachment, penetration, uncoating, replication, assembly, and release of respiratory viruses. Further analysis of the potential phytochemicals in treating SARS-CoV-2 in clinical trials is warranted.
