Vitamin D3 Protects against Diabetic Retinopathy by Inhibiting High-Glucose-Induced Activation of the ROS/TXNIP/NLRP3 Inflammasome Pathway

Abstract: Purpose This study aimed to evaluate the mechanisms underlying the effects of 1,25-dihydroxyvitamin D (vitamin D3) on diabetes-induced retinal vascular damage and retinal vascular endothelial cell apoptosis. Methods Diabetic and control rats were randomly assigned to receive vitamin D3 or vehicle for 6 months. Additionally, human retinal microvascular endothelial cells (HRMECs) were incubated in normal or high-glucose medium with or without vitamin D3. Morphological changes in retinal tissues and retinal vascu… Show more

“…153 In a streptozotocin-induced T1DM model, NLRP3 upregulation is observed in the retina, 148,149 which may contribute to DR by inducing vascular damage and cell death. 154 NLRP1, also known as NALP1, inflammasome is the first member of the NLR family to be discovered, playing an essential role in inflammatory responses. Knockout of NLRP1 protects against diabetesinduced proinflammatory cytokine and VEGFA expression and thereby attenuates DR. 151 VEGFA-transgenic mice, which develop retinal neovascularization, express high levels of NLRP3 and IL-1β in retinas.…”

“…However, the target relation between miR‐20b‐3p and TXNIP has not been uncovered yet. Furthermore, inflammasomes act as an intracellular machinery to initiate inflammatory response to many danger signals, and the relation between TXNIP and nucleotide‐binding oligomerization domain‐like receptors (NLR) family pyrin domain containing 3 (NLRP3) inflammasomes in DR has been unveiled by several literatures …”

“…The TXNIP/NLRP3 in ammasome plays a key role in the pathogenesis of various diseases [30,32,34,35]. Chen W et al [9] have indicated that minocycline improves diabetic retinopathy by inhibiting the TXNIP/NLRP3 in ammasome pathway, and Vitamin D3 attenuates diabetic retinopathy by inhibiting high-glucose-induced ROS/TXNIP/NLRP3 in ammasome pathway [36]. In the present study, we found that the TXNIP/NLRP3 in ammasome components were upregulated in EAE mice, and bixin remarkably downregulated the expression of TXNIP and NLRP3 at both mRNA and protein levels.…”
