Vitamin A and vitamin D3 protect the visual apparatus during the development of dopamine-2 receptor knockout mouse model of Parkinsonism

Abstract: Background Dopamine-related movement disorders are associated with a loss of visual acuity. Studies have shown that chemical stimulation of the Vitamin D3 receptor (VDR) ameliorates movement disorders; however, the chemical stimulation is not effective when there is a deficiency of Vitamin A in the cells. In the study, we examine the role of VDR and its interplay with vitamin A in impaired visual function in the dopamine deficit model. Methods… Show more

“…This suggests that our EPS model replicates the pathognomonic traits of PD, demonstrating good construct validity. Post-mortem examinations of Parkinson's disease patients' brains revealed a negative correlation between neuronal death and the number of activated astrocytes [36,37]. Therefore, the substantial increase in the expression of activated astrocytes indicates signi cant neurodegeneration in the corticobasal loop resulting from prolonged inhibition of the dopamine-2 receptor used in developing the EPS model.…”

“…A model of EPS was developed using 10 mg/kg body weight (BW) of a dopamine-2 receptor blocker, in line with an earlier protocol [15,37]. 800 IU of vitamin D3 and 100 IU of vitamin A were used for vitamin D3 and vitamin A treatments, respectively.…”
