Visualization and tracking of tumour extracellular vesicle delivery and RNA translation using multiplexed reporters

Abstract: Accurate spatiotemporal assessment of extracellular vesicle (EV) delivery and cargo RNA translation requires specific and robust live-cell imaging technologies. Here we engineer optical reporters to label multiple EV populations for visualization and tracking of tumor EV release, uptake and exchange between cell populations both in culture and in vivo. Enhanced green fluorescence protein (EGFP) and tandem dimer Tomato (tdTomato) were fused at NH2-termini with a palmitoylation signal (PalmGFP, PalmtdTomato) for… Show more

“…6 To confirm that FN is located on the exosome surface as would be expected, we used a dot blot assay that can distinguish EV surface proteins from lumen proteins based on permeabilization of EVs in 0.1% Tween-20. 33,34 The tetraspanin CD63 was used as a positive control, as it is a well-characterized exosomal surface protein. Using an antibody to the large extracellular loop, CD63 was detected in both detergent-free and -containing conditions (Fig.…”

“…In tracing the target cells of EVs, transmembrane proteins fused to luciferase or fluorescent proteins in EVs can be used as tracers, and the luciferase or fluorescent activity of distant cells can be measured to trace the effects of EVs 42, 111…”

“…Lai et al designed a practical tool, EVs containing fluorescently labeled mRNA to confirm the internalization of EVs by measuring the signal 42. Another effective and more proven method utilizes the Cre–LoxP system, in which transgenic mice express the LacZ reporter gene with a STOP sequence, which can be excised by Cre to subsequently exhibit LacZ expression after transfection with EVs containing Cre mRNA 113.…”

“…More studies addressing this issue are needed that could discover the tumor-specific targeting of these exosomes. Quick biodistribution of exosomes applied systemically and their elimination in 1-6 hours after administration [33][34][35] suggests that AMF exposition should be applied shortly after the magnetoexosome application.…”
