Visualisation of Multiple Tight Junctional Complexes in Human Airway Epithelial Cells

Abstract: BackgroundApically located tight junctions in airway epithelium perform a fundamental role in controlling macromolecule migration through paracellular spaces. Alterations in their expression may lead to disruptions in barrier integrity, which subsequently facilitates entry of potential bacterial and other pathogens into the host. Furthermore, there is emerging evidence that the barrier integrity of the airway in certain airway inflammatory diseases may be altered. However, there is little consensus on the way … Show more

“…The formation of junctional-complexes (which include tight junctions, adherens junctions and desmosomes) between cells creates a physicochemical barrier against inhaled particulates and prevent the penetration of microorganisms and chemicals into the interstitial compartment 3 , 5 , 59 . Tight junctions are involved in the regulation of solute and ion transport across epithelia and are localised at the apicolateral border of individual airway epithelial cells 3 , 5 , 60 .…”

“…Concentrations of the tracers in the basolateral medium are measured and an apparent permeability coefficient ( P app ) of the epithelium calculated (Strengert and Knaus 2011 ). Besides these quantitative assays, confocal microscopy also has been used as a visual tool for studying the dynamics of TJs (Buckley et al 2018 ). Depending on the purpose of the studies, these methods can be used in combination to comprehensively assess the tissue barrier properties of the ALI cultures under experimental conditions.…”

“…Delivery of prepared phages could also be tested on AEC ALI cultures to capture a more comprehensive pre and post application reaction of the host mammalian cells ( Buckley et al, 2018 ). While topical and/or intravenous applications of delivery have been effective, they are not ideal for pulmonary infections, particularly in pediatrics.…”
