Visualisation of direct interaction of MDA5 and the dsRNA replicative intermediate form of positive strand RNA viruses

Abstract: SummaryThe innate immune system is a vital part of the body's defences against viral pathogens. The proteins retinoic acid-inducible gene-I (RIG-I) and melanoma differentiation associated gene 5 (MDA5) function as cytoplasmic pattern recognition receptors that are involved in the elimination of actively replicating RNA viruses. Their location and their differential responses to RNA viruses emphasises the complexity of the innate detection system. Despite the wealth of information on the types of RNA that trigg… Show more

“…The monoclonal anti-dsRNA J2 antibody binds dsRNAs at least 40 bp long in a sequence-independent fashion (12)(13)(14). The J2 dsRNA monoclonal antibody was previously used to detect viral dsRNA by dsRNA immunoblotting, immunohistochemistry, ELISA, and immunoprecipitation in eukaryotic cells (13)(14)(15)(16)(17)(18)(19)(20). In this study, we used the J2 dsRNA antibody in an immunoprecipitation assay to enrich dsRNA from total RNA extracted from E. coli.…”

“…46 The viral ligand for MDA5 is long higher order dsRNA or dsRNA replication intermediates. 21; 47; 48; 49; 50 Interestingly, during infection of West Nile virus, an RNA virus also in the Flaviviridae, both RIG-I and MDA5 contribute to the antiviral response. In this case, RIG-I is important early after infection, while MDA5 is important for signaling during later times of infection.…”

“…MDA5 ligands are less well defined, but MDA5 responds more specifically to long dsRNA that contain branched or higher order structures [6] and senses the absence of 2'-O-methylation at the 5′ cap structure of viral RNA [7]. Direct binding of viral RNA to MDA5 was not yet demonstrated, but colocalization of MDA5 and the replicative dsRNA of enteroviruses was observed using fluorescent imaging [8]. …”
