Virulotyping and Antimicrobial Resistance Typing ofSalmonella entericaSerovars Relevant to Human Health in Europe

Abstract: The combination of virulence gene and antimicrobial resistance gene typing using DNA arrays is a recently developed genomics-based approach to bacterial molecular epidemiology. We have now applied this technology to 523 Salmonella enterica subsp. enterica strains collected from various host sources and public health and veterinary institutes across nine European countries. The strain set included the five predominant Salmonella serovars isolated in Europe (Enteritidis, Typhimurium, Infantis, Virchow, and Hadar… Show more

“…Escherichia coli ATCC 25922 was used as a control strain. Virulence-associated genes pefA, spvC, sirA, gipA, SEN1417, trhH, and prot6e, all of which have been variably detected in the S. Enteritidis serotype (Pan et al 2009, Huehn et al 2010, were used as target sequences for the PCRbased virulotyping. DNA extraction was performed by the High Pure PCR Template preparation Kit Ò (Roche, Mannheim, Germany) following the manufacturer's recommendations.…”

“…Currently, alternative molecular typing methods such as multilocus sequence typing (MLST) and virulotyping [1,[14][15] are used to characterize many Salmonella enterica serovars. Multilocus sequence typing [16] has been increasingly recognized as a method of choice for genotyping a number of bacterial pathogens [17][18], including Salmonella [1,[19][20][21], and has been used successfully in epidemiological studies and outbreak investigations [17,22].…”

“…Virulence genes have been screened using simple [25] or multiplex PCR [26] to determine the distribution of these genes among Salmonella enterica strains [25,27] and to genotype many Salmonella serovars [15,[28][29].…”

“…These ''epidemic'' monophasic isolates have been found to lack fljB, a gene that encodes phase 2 flagella Huehn et al, 2010). Monophasic members of Salmonella predominantly belong to two main phage types, DT193 and DT104, and pigs are considered the predominant source Huehn et al, 2010;Wasyl & Hoszowski, 2012).…”
