Introduction: Several Escherichia coli pathotypes have been reported in Thailand; however, information on enteroaggregative heat-stable enterotoxin 1 (EAST1)-carrying E. coli (EAST1-EC) is insufficient. Previous reports show that consumption of raw meats causes diarrheagenic E. coli infections. In this study, we investigated the seroprevalence and genetic relationship of EAST1-EC from clinical and raw meat samples. Methodology: Diarrheal patients and raw meat samples were investigated for the presence of EAST1-EC by performing polymerase chain reaction (PCR) to detect astA. Serotyping, antimicrobial susceptibility tests, and PCR-based phylogenetic group assay were performed. Molecular epidemiology of E. coli strains from clinical and raw meat samples was determined using repetitive element-PCR typing, BOX-PCR, and ERIC2-PCR. Results: Results showed that 11.2% (17/152) of clinical samples and 53.3% (16/30) of raw meat samples had EAST1-EC. In all, 24 and 36 EAST1-EC strains were successfully isolated from 17 clinical and 16 raw meat samples, respectively. These strains had astA but did not possess the indicative genes of other E. coli pathotypes and were therefore classified as EAST1-EC. Most of these strains were multidrug resistant and were classified into nine serogroups. Molecular genotyping showed identical DNA fingerprint among EAST1-EC serotype O15 strains from clinical and raw chicken samples, suggesting that they were derived from the same bacterial clone. Conclusions: Our results indicated a high prevalence of multidrug-resistant EAST1-EC strains in clinical and environmental samples in Thailand belonging to nine serogroups. Moreover, the study highlighted the close association between infections caused by EAST1-EC serotype O15 and raw meat consumption.

A total of 202 stool samples from human, cats and dogs were examined for detection of H.pylori and E.coli. For isolation of H.pylori, 150 stool samples were cultivated on Dent , s medium (modified skirrow , s medium) containing selective Dent , s antibiotic supplement. while the remaining 52 samples were cultivated on MacConkey, Eosine methylen blue media in attempt to isolate other associated bacteria. Colonies grossly similar to H.pylori were tested for catalase, oxidase and urease test. All human samples were tested serologically by using rapid stool antigen test (SAT). PCR assay was performed on bacterial isolates and directly from stool samples by extraction of DNA from stool by boiling method. Positive cultures were subjected to serology testing and PCR. 45samples from humans gave positive results with rapid H.pylori stool antigen test (HPSA). Results revealed that H.pylori was detected in tested samples from human and cat at a rate of 52.8%, 33.3%, respectively and could not be detected in dog. Also detected serologically at percentage of 84.9% in human stool samples. H.pylori could not be detected by PCR, while the other 52 samples were found positive for Escherichia coli, isolation at a percentage of 48%. Nine serogroups of E.coli (O1, O20, O55, O86, O114, O125, and O146) were detected in 9 samples (3 samples from each species). All serotypes were positive for Iss and Fim genes. From all stool samples subjected for isolation of E.coli we obtained 15 isolates which were introduced to PCR. 14 isolates were positive for E.coli genus (phoA gene). 9 isolates were used for detection of virulence genes (Iss, Fim, and Tsh genes). 8 isolates were positive for (Iss and Fim) gene. None was positive for (Tsh) gene. We conclude that H.pylori is very difficult to culture due to the fastidious nature of the organism and its transport and culture requirements. Also H.pylori is the main species of Helicobacter present in human. While dogs and cats harbour many gastric Helicobacter like organisms (GHLOs) and E.coli isolated from human, dogs and cats share the same characters and virulence genes.

Avian pathogenic Escherichia coli (APEC) causes colibacillosis, which is one of the main causes of economic losses in the poultry industry worldwide. This disease occurs only when the E. coli infecting strain presents virulence factors (encoded by specific genes) that enable the adhesion and proliferation in the host organism. Herin, 15 E. coli strains of different serogroups isolated from birds with colibacillosis were assigned to their phylogenetic groups and analyzed for the occurrence of 11 virulence associated genes. phylogenetic typing showed that group B2 was the largest (33.3%, 5/15) followed by group B1 (26.7%, 4/15), groups A and D were similar in size (22.1%, 19/86). The virulence profiles showed that ompA was found in most isolates 14/15 (93.3%). The iss gene was found in13/15 (86.6%). Followed by traT and iutA genes which were found in12/15 (80%). cvaC, stx2 and tsh genes were present in 9/15(60%), 7/15(46%) and of the isolates, respectively. Only one isolate gave positive amplification for stx1 and ibeA genes each. The hly gene was not encountered in any of the tested isolates.
