Virion-Associated Restriction Endonucleases of Chloroviruses

Abstract: Chloroviruses are large, double-stranded-DNA, plaque-forming viruses that infect certain eukaryotic chlorella-like green algae. The prototype of the genus is Paramecium bursaria chlorella virus 1 (PBCV-1). Chlorovirus genomes contain various amounts of methylated nucleotides due to virus-encoded DNA methyltransferases (MTases); about 25% of the MTases are associated with companion DNA site-specific (restriction) endonucleases (REases). These enzymes constitute virally encoded restriction-modification (R/M) sys… Show more

“…ATCV-1 was grown in C. heliozoae SAG 3.83 cells, purified by successive rounds of gradient centrifugation and 1% Triton X-100 and proteinase K treatments as previously described (17), with some modifications. Because of the sensitivity of ATCV-1 to sucrose, two iodixanol gradient centrifugations were substituted for the sucrose gradients.…”

“…Consequently, the ultrapurification procedure resulted in two proteinase treatments and three iodixanol gradient centrifugations. Chlorella viruses PBCV-1 and CVM-1 were grown in Chlorella variabilis strain NC64A and M. conductrix Pbi, respectively, and purified as described previously (17). Stock preparations were maintained in virus stabilization buffer (50 mM TrisHCl, 10 mM MgCl 2 , pH 7.8) at 1 ϫ 10 11 PFU/ml, which was changed to phosphate-buffered saline (PBS) at 1 ϫ 10 10 PFU/ml at the time of use.…”

“…The growth of the host Chlorella strain NC64A on bold basal medium (BBM) modified by the addition of 0.5% sucrose and 0.1% peptone (MBBM) and the production and purification of viruses PBCV-1 and NY-2A have been previously described elsewhere (1,43).…”

“…Pulse field gel electrophoresis studies were performed according to Agarkova et al (2006). Chromosomal DNAs were separated in a CHEF-DR II (Bio-Rad) unit in a 0.8% agarose gel.…”
