An antimicrobial substance producing strain designated as A52 was isolated from a marine sediment sample and identified as Bacillus sp., based on 16S rRNA gene sequence analysis. The ANI and dDDH analysis of the genome sequence displayed high identity with two strains of B. subtilis sub sp. subtilis. Strain A52 yielded two antimicrobial peptides (AMPs) that differed in activity spectrum. MALDI mass spectrometry analysis of HPLC purified fractions revealed mass of peptides as 3881.6 and 1061.9 Da. The antiSMASH analysis of genome sequence unraveled presence of identical biosynthetic cluster involved in production of sublancin from B. subtilis sub sp. subtilis strain 168, which yielded peptide with identical mass. The low molecular weight peptide is found to be a cyclic lipopeptide containing C 16 β-hydroxy fatty acid that resembled surfactin-like group of biosurfactants. However, it differed in fatty acid composition and antimicrobial spectrum in comparison to other surfactins produced by strains of B. subtilis. It exhibited broad spectrum antibacterial activity, inhibited growth of pathogenic strains of Candida and filamentous fungi. Further, it exhibited hemolytic activity, but did not show phytotoxic effect in seed germination experiment. The emulgel formulation of surfactin-like lipopeptide showed antimicrobial activity in vitro and did not show any irritation effects in animal studies using BALB/c mice. Moreover, surfactin-like lipopeptide displayed synergistic activity with fluconazole against Candida, indicating its potential for external therapeutic applications.

Microbial communities from various environments have been studied in the quest for new natural products with a broad range of applications in medicine and biotechnology. We employed an enrichment method and genome mining tools to examine the biosynthetic potential of microbial communities in the sediments of a coastal sinkhole within the karst ecosystem of the Yucatán Peninsula, Mexico. Our investigation led to the detection of 203 biosynthetic gene clusters (BGCs) and 55 secondary metabolites (SMs) within 35 high‐quality metagenome‐assembled genomes (MAGs) derived from these subcommunities. The most abundant types of BGCs were Terpene, Nonribosomal peptide‐synthetase, and Type III polyketide synthase. Some of the in silico identified BGCs and SMs have been previously reported to exhibit biological activities against pathogenic bacteria and fungi. Others could play significant roles in the sinkhole ecosystem, such as iron solubilization and osmotic stress protection. Interestingly, 75% of the BGCs showed no sequence homology with bacterial BGCs previously reported in the MiBIG database. This suggests that the microbial communities in this environment could be an untapped source of genes encoding novel specialized compounds. The majority of the BGCs were identified in pathways found in the genus Virgibacillus, followed by Sporosarcina, Siminovitchia, Rhodococcus, and Halomonas. The latter, along with Paraclostridium and Lysinibacillus, had the highest number of identified BGC types. This study offers fresh insights into the potential ecological role of SMs from sediment microbial communities in an unexplored environment, underscoring their value as a source of novel natural products.

Neglected invertebrates, marine flatworms, have attracted global research interest due to their biological and chemical potential properties. The marine flatworms (Turbellaria), Phylum Platyhelminthes, belong to the Polycladida group. There are about 3000 species of free-living flatworms that make a living by hunting and eating other animals. A marine flatworm Acanthozoon sp. was used in this study due to its abundant presence in the site location. Staphylococcus epidermidis is an opportunistic pathogenic bacterium that was previously considered a harmless skin disease bacterium. This species is now considered to be in the first rank among the causative agents of nosocomial infection, specifically in the form of infections of the urinary tract, respiratory tract, and surgical site wounds. The aims of this study were to explore the biological diversity of marine flatworm-associated bacteria with antipathogenic properties and to detect the presence of polyketide synthase (PKS) and nonribosomal peptide synthetase NRPS gene clusters through a molecular approach. Recent studies have shown that S. epidermidis undergoes functional changes in the pro-inflammatory peptide family so that it has functions in immune evasion and biofilm development. Therefore, the search for new antimicrobial compounds is urgently needed due to the limited choice of antibiotic use. In the preliminary screening by overlay test, 7 out of 17 (41.2%) isolates showed antibacterial activities. These isolates were reselected and their activity confirmed by using plug agar and disk-diffusion methods. The FA02, FA03, FA05, FA07, FA13, FA16, and FA17 isolates demonstrated their inhibitory activities consistently against the causative agent of nosocomial infection S. epidermidis. Based on the morphological and 16S rRNA partial sequencing analysis, these isolates were closely related to the genus Virgibacillus, Brevibacterium, Alcanivorax, and Vibrio. None of these seven antibacterial strains possesses PKS-I and PKS-II, except NRPS genes for Virgibacillus salarius strain FA02, V. salarius strain FA16, and V. salarius strain FA17. The results of this study showed that bacteria associated with marine flatworms have future potential as a source of promising natural products for the development of antibiotics.
