VirF Relieves the Transcriptional Attenuation of the Virulence Gene icsA of Shigella flexneri Affecting the icsA mRNA–RnaG Complex Formation

Abstract: VirF is the master activator of virulence genes of Shigella and its expression is required for the invasion of the human intestinal mucosa by pathogenic bacteria. VirF was shown to directly activate the transcription of virB and icsA, which encode two essential proteins involved in the pathogenicity process, by binding their promoter regions. In this study, we demonstrate by band shift, enzymatic probing and cross-linking experiments that VirF, in addition to DNA, can also bind the icsA transcript and RnaG, an… Show more

“…Interestingly, VirF may be acting through the displacement of H-NS around the +1 of transcription and in the coding sequence of icsA [114], while in the case of virB, it more classically binds to the promoter [105,108]. On top of this antisilencing effect on transcription, VirF was recently suggested to regulate the expression of icsA by directly binding its mRNA and its noncoding antisense regulator RnaG [115].…”

“…The pulldown assay was performed as described previously ( 51 ). The biotinylated RNA probe targeting ssaV was generated using the Angle gene (Nanjing, Jiangsu, China) and incubated with streptavidin magnetic beads (Thermo Fisher Scientific, Waltham, MA, USA) at 4°C overnight.…”

“…In contrast, H-NS silences icsA at 30 °C, whereas at 37 °C VirF relieves H-NS-mediated silencing, thereby upregulating icsA transcription [ 77 ]. At 37 °C, VirF may also repress the expression of rnaG [ 77 ] and stabilize icsA mRNA by disrupting the transcription attenuation duplex the latter can form with RnaG [ 78 ]. Taken together, these studies suggest that VirF dual-activates icsA through the release of both H-NS transcriptional repression and RnaG attenuation of its mRNA.…”
