Viral hemagglutinin augments peptide‐specific cytotoxic T cell responses

Abstract: In attempt to increase the induction of peptide-specific cytolytic T cells (CTL) we investigated the effect of the Newcastle disease virus (NDV) hemagglutinin-neuraminidase (HN) gene product on the activation of peptide-specific CTL. Spleen cells of CH3 mice immunized against the influenza nucleoprotein peptide 50-63 (NP 50-63) were restimulated in vitro (i) with peptide-pulsed syngeneic fibroblast cells (Ltk-) as antigen-presenting cells, which were in addition (ii) infected with NDV or (iii) stably transfect… Show more

“…10,11 In a murine tumor model, immunization with ATV-NDV did not change the specificity of the T-cell-mediated antitumor response, 12 although it did selectively increase the frequency of tumor-specific CTL precursors 12 and potentiate tumor-specific CTL activity via the induction of type I interferons (IFNs) 16 and via the HN molecule. 15,17 There was no nonspecific mitogenic or adjuvant effect of NDV. 17 Virus potentiation of tumor vaccine T-cell stimulatory capacity required cell surface binding but not infection.…”

“…25 Immunostimulatory properties of the hemagglutininneuraminidase (HN) of the Newcastle disease virus (NDV) have been demonstrated in vitro. 26,27 Mouse fibroblasts transfected with HN or infected with NDV have been shown to stimulate specific T-cell proliferation with greater efficacy than untreated fibroblasts. 26 A major effect of NDV infection is the induction of IFN-a/b secretion, which can be mediated either through TLRs or by RIG-I-dependent signaling.…”
